MiR-99a Inhibits Keratinocyte Proliferation by Targeting Frizzled-5(FZD5)/FZD8 in Psoriasis

【Objective】The present study is aimed to investigate the molecular mechanism of miR-99a targeting FZD5/FZD8 to inhibit the proliferation of keratinocytes in psoriasis. 【Methods】The expression levels of miR-99a, FZD5/FZD8, β-catenin and CyclinD1 in tissues and cell lines were detected by QPCR and Western Blot. The proliferation of keratinocytes was detected by MTT and BrdU assays. The target gene of miR-99a was verified by dual luciferase reporter gene assay. 【Results】The expression of miR-99a was significantly down-regulated in psoriatic lesions. Overexpressed miR-99a downregulated the expression of FZD5/FZD8, β-catenin and CyclinD1, and inhibited keratinocyte proliferation as well. On the other hand, the inhibition of miR-99a showed upregulated FZD5/FZD8, β-catenin and CyclinD1 expression, while keratinocyte proliferation got enhanced. It was found that miR-99a targeted the regulation of FZD5 and FZD8. Overexpression of FZD5/FZD8 restored the inhibitory effect of miR-99a on β-catenin and CyclinD1 proteins. The expression of miR-99a and FZD5/FZD8 was inversely correlated. 【Conclusion】 MiR-99 can target FZD5/FZD8 gene to down-regulate their expression and inhibit keratinocyte proliferation through β-catenin/CyclinD1 signaling.

Experimental Study on the Inhibition of Magnolol on Biological Activities of Human Breast Cancer Cell Lines in Vitro

【Objective】To investigate the effects of Magnolia (Magnolol, Mag) that inhibited the biological activity of different breast cancer cell lines in vitro. 【Methods】 Human breast cancer high metastatic cell lines MCF-7 and MDA-MB-231 were used in the study. Different concentrations of Mag solution were added to the culture medium to observe the rate of apoptosis, invasion, migration and adhesion capacity. Then, the effects of Mag on biological activity of breast cancer cell line were evaluated. 【Results】Data showed that Mag inhibited the rate of apoptosis, invasion, metastasis and adhesion ability in two breast cancer cell lines (P<0.05), and that with the increase of Mag concentration, the inhibitory activity on the biological activity was increased (P<0.05). 【Conclusion】Mag can inhibit the biological activities of breast cancer cell lines by inhibiting the apoptotic rate, the ability of invasion, and the adhesion and migration of breast cancer cell lines.It has an important value in the treatment and prevention of breast cancer recurrence or metastasis in clinic.

Factors Influencing Prognosis of Patients with Acute Paraquat Poisoning within 24 Hours of Admission

【Objective】To investigate the risk factors of acute paraquat poisoning in patients who were hospitalized within 24 hours. 【Methods】 A total of 123 patients with acute paraquat poisoning were divided into survival group (n=46) and death group (n=77) from Jan. 2009 to Aug. All patients were recorded and analyzed for general information (gender, age, oral dose of paraquat, time to gastric lavage and perfusion, number of perfusions), concentration of paraquat at admission, worst APACHE Ⅱ score within 24 hours, blood gas analysis, blood Routine, urine, liver and kidney function, myocardial enzymes, electrolytes and other biochemical indicators. The differences of the indexes between the two groups were compared and the prognostic risk factors were analyzed. 【Results】 There was no significant difference in the age and the difference between the survival group and the death group (P>0.05). The survival dose and the concentration of paraquat in the surviving group were lower than those in the death group (P<0.05), but there was no significant difference between perfusion time and perfusion time. The levels of AST(Aspartate aminotransferase), Tbil(Serum total bilirubin), BUN(Urea nitrogen) and CK(Creatine kinase) in the survivors group and the death group were significantly higher than those in the survivors group (P<0.05); the levels of leukocyte, ALT(Alanine aminotransferase) and Cr(Creatinine) in the dead group were significantly higher than those in the survival group (P>0.05). Effects of Abnormal Lipid Metabolismin of Early Pregnancy on the Incidence Rate of Gestational Diabetes Mellitus

【Objective】To explore the effects of abnormal lipid metabolism in early pregnancy on the incidence rate of gestational diabetes mellitus (GDM).【Methods】A total of 120 pregnant women with abnormal lipid metabolism in the early pregnancy were selected as the observation group, and other 120 pregnant women with normal lipid metabolism in the early pregnancy were selected as the control group. The levels of serum lipid metabolism indexes cholesterol(TC), triglyceride (TG), low density lipoprotein cholesterol(LDL-C), high density lipoprotein cholesterol (HDL-C), adipocytokine leptin, free fatty acid, blood glucose and insulin resistance indexes fasting blood glucose (FBG), glycosylated hemoglobin (HbAlc), fasting membrane insulin (FINS), insulin resistance index (HOMA-IR) were measured in the two groups. The correlation of TG, leptin and blood glucose and insulin resistance indexes was analyzed, and the incidence rate of GDM, pregnancy outcome, fetal and related complications were analyzed in two groups.【Results】Compared with the control group, the levels of TG, TC and LDL-C in the observation group were significantly higher while the level of HDL-C was significantly lower (P<0.05), and leptin and free fatty acid were significantly higher (P<0.01). At the same time, the levels of blood glucose indexes FBG, HbAlc and HOMA-IR were significantly higher in the observation group, and the insulin resistance index FINS was significantly lower (P<0.01). Univariate correlation analysis showed that the levels of TG and leptin were positively correlated with FBG, HbAlc and HOMA IR, but negatively correlated with FINS (P<0.05). The incidence rate of GDM was 35% in the observation group, which was significantly higher than that in the control group with16.67%% (P<0.05). Compared with the control group, the spontaneous labor rate of the observation group was significantly lower, and the cesarean section rate was significantly higher (P<0.05). And the incidence rates of fetal distress and macrosomia in the observation group were significantly higher than those of the control group (P<0.05).【Conclusion】Early pregnancy lipid metabolism abnormalities which contains the abnormal lipids, adipocytokine levelscan significantly increase the incidence rate of GDM, and affect the pregnancy outcome of pregnant women and fetal birth.

Anti-Proliferation and Pro-apoptosis Effects of Decitabine Combined with Idarubicin on AML Cells Harboring Mutant DNMT3A

【Objective】To evaluate the antiproliferation and pro-apoptotic effects of decitabine (DAC) combined with idarubicin (IDA) on OCI-AML3 cells harboring mutant DNMT3A.【Methods】OCI-AML3 cells were treated with different concentrations of IDA singly or combined with 2μM DAC for 48 h. Then, MTT and Annexin-V/PI were used to examine the proliferation and apoptosis of OCI-AML3 cells.【Results】IDA at relatively low concentrations (≤40nM) did not inhibit proliferation of OCI-AML3 cells significantly, but promoted proliferation inhibition remarkably when combined with 2μM DAC (P<0.01). DAC combined with IDA induced higher apoptotic rate compared to DAC or IDA as a single agent (P<0.001).【Conclusion】AML cells harboring mutant DNMT3A are primarily resistant to IDA at relatively low concentrations, while DAC can increase the susceptibility to IDA.

Effect of Noninvasive Bi-Level Positive Airway Pressure Combined with Respiratory Stimulant Treatment on Chronic Obstructive Pulmonary Disease (COPD) with Pulmonary Encephalopathy

【Objective】To investigate the changes of the inflammation factor levels and the clinical effect of noninvasive bi-level positive airway pressure (BiPAP) combined with respiratory stimulant treatment on COPD patients with mild to moderate pulmonary encephalopathy. 【Methods】A total of 80 patients with COPD and mild to moderate pulmonary encephalopathy at the Central Hospital of Jiangjin District between December 2013 and December 2015 were randomly divided into the observation group (n=40) and the control group (n=40). Patients in both groups received routine supportive treatments in addition to BiPAP ventilation while the observation group was treated with respiratory stimulant. A variety of the indicators (PaO

in the observation group improved significantly more than those in the control group (P<0.05). PaCO

, RR and HR were significantly lower in the observation group than in the control group, and aeration time, consciousness recovery time, hospital stay and tracheal intubation rate were significantly shorter (all P<0.05). The levels of IL-6, IL-8 and IL-10 in the observation group were significantly lower than those in the control group (all P<0.05).The effective rate of the observation group was 90.0%, which was higher than that of the control group (47.5%); it showed statistical significance (P<0.05) . 【Conclusion】Application of BiPAP combined with respiratory stimulants is an effective treatment for COPD patients with pulmonary encephalopathy. It can reduce the tracheal intubation rate and shorten the hospitalization time.

Effects of Ganmai Dazao Decoction on Learning, Memory and Hippocampus Synaptic Plasticity of Chronic Stress Depression Model Rats

【Objective】To investigate the effects of Ganmai Dazao Decoction on learning, memory and hippocampus synaptic plasticity in chronic depression model rats. 【Methods】Wistar rats were randomly divided into 4 groups: the control group, he depression model group, the fluoxetine group (5.4mg/kg) and the Ganmai Dazao Decoction (3.84 g/kg) group. Depression models were established by chronic mild unpredictable stress and solitude. The fluoxetine hydrochloride and Ganmai Dazao Decoction group were intra-gastrically administrated (ig) with corresponding drugs once a day for 21 days, and the depression model group was intraperitoneally injected with water. Then, we observed the effects of water maze test in model rats with Ganmai Dazao Decoction. The expression of SYN I (synaptophysin I), SYNA (synaptophysin), and BDNF (brain-derived neurotrophic factor) were detected by the immunohistochemistry and Western-Blotting methods. 【Results】Compared to the control group, the horizontal and vertical activity scores of the model group decreased significantly (P<0.01). However, the escape latency (EL) and target quadrant latency increased significantly (P<0.05), and the expression of SYN I, SYNA, BDNF were reduced dramatically as well. Compared to the outcomes of the model group, Ganmai Dazao Decoction reduced the EL (P<0.05), shortened the target quadrant latency (P<0.05) and increased the expression of hippocampal SYN I, SYNA and BDNF(P<0.01 or P<0.05) of the model rats.【Conclusion】Ganmai Dazao Decoction can significantly improve the learning and memory abilities in chronic depression model rats, which may play a role in the promotion of hippocampal synaptic plasticity.

Mechanism and Effect Ossotide Injection on Osteoporosis induced by Retinoic Acid in Rats

【Objective】 To explore the mechanism and effect of Ossotide injection on osteoporosis induced by retinoic acid in rats. 【Methods】 Fifty SD rats were randomly divided into normal control group, model control group, Ossotide injection low, medium and high groups. In addition to normal control group, and the other four groups were intraperitoneally injected with 70 mg/kg retinoic acid for 14 days. Ossotide injection low, medium and high groups were intragastrically administrated with 100, 200, 400 μg/(kg·d) Ossotide injection, the normal control group and the model control group were given equal amount of normal saline for 4 weeks. The rat bone mass, bone mineral density was observed and recorded. The level of calcium, phosphorus and alkaline phosphatase (ALP) in serum was detected by automatic biochemical analyzer method. The level of serum tartrate resistant acid phosphatase (TRACP) activity and osteocalcin content was detected by kit. 【Results】Compared with normal group, the level of calcium and phosphorus in serum, bone mass, bone mineral density was decreased (P<0.01), the activity of ALP and TRACP was increased (P<0.01), the level of osteocalcin was increased in model group (P<0.01). Compared with model control group, the level of calcium and phosphorus, bone mass, bone mineral density in serum was increased (P<0.01), the activity of ALP and TRACP was decreased (P<0.01), the level of osteocalcin was decreased in Ossotide injection low, medium and high group (P<0.01). 【Conclusion】Ossotide injection could treat retinoic acid induced osteoporosis in rats.

Significance of Survivin, MMP-2 and MMP-9 Expression in Thyroid Carcinoma

【Objective】To investigate the expression of Survivin, matrix metalloproteinase-2 (MMP-2) and MMP-9 in thyroid carcinoma, and to explore its significance. 【Methods】The expression of Survivin, MMP-2 and MMP-9 in 86 cases of thyroid carcinoma, 40 cases of benign thyroid tissue, and 86 cases of adjacent normal tissues were detected by the immunohistochemical method. The relationship between Survivin, MMP-2 and MMP-9 and clinical pathological parameters was analyzed. 【Results】 The positive rate of Survivin, MMP-2 and MMP-9 in adjacent normal tissues, benign thyroid tissue and thyroid carcinoma increased in that order . The expression of Survivin, MMP-2, and MMP-9 in thyroid carcinoma was significantly higher than that in the benign thyroid tissues and adjacent normal tissues (P<0.001). The positive rate of Survivin, MMP-2 and MMP-9 in Ⅲ~Ⅳ stage was higher than that in theⅠ~Ⅱ stage (P<0.05). The positive rate of Survivin, MMP-2, and MMP-9 in lymph node metastasis cases was higher than that in in cases without lymph node metastasis (P<0.05). The positive rate of survivin, MMP-2, and MMP-9 in thyroid carcinoma wasn't related to age, gender and tumor diameter. The expression of Survivin, MMP-2, and MMP-9 in thyroid carcinoma was positively correlated with each other. 【Conclusion】Survivin, MMP-2 and MMP-9 genes play an important role in the occurrence and development of thyroid carcinoma. The combined detection of the three might be a diagnostic index for the diagnosis and treatment of thyroid cancer.

Clinical Effect and Influencing Factors of Mifepristone in the Conservative Treatment of Peri Menopausal Uterine Fibroids

【Objective】 To investigate the clinical effect of mifepristone in the treatment of uterine fibroids and its influencing factors. 【Methods】 Forty patients with myoma of uterus treated in our hospital from December 2010 to June 2016 were selected; Mifepristone was used to compare the volume change of hysteromyoma before and after treatment, and the factors influencing the effect of mifepristone treatment were analyzed.【Results】 The volume of myoma of uterus decreased significantly after treatment (P<0.05); The effect of mifepristone was poor in patients with myoma of uterus larger than 3 cm, ER receptor negative, uterine fibroids >6 months and the fibroids located under the serosa(P<0.05). 【Conclusion】 Mifepristone is effective in the treatment of uterine fibroids; ER receptor, myoma size and course of disease are the main factors that influence the effect of mifepristone treatment.

A Comparative Study on the Effects of Three Surgical Methods for Treating Middle and Lower Esophageal Cancer and the Changes of Stress Indexes

【Objective】 To observe the effects of three surgical methods for treating middle and lower esophageal cancer and the changes of stress indexes. 【Methods】Eighty patients with middle and lower esophageal cancer were divided into three groups. 25 cases in group A were treated by left thoracic single-port resection, while 28 cases in group B were treated by right thoracic and abdominal resection, and 27 cases in group C were treated by left neck, right thoracic and median abdominal resection. The perioperative indexes and the number of dissected lymph nodes were compared between the groups, the white blood cell count (WBC), serum free triiodothyronine (FT3), free thyroxine (FT4), CRP, cortisol, postoperative complications and 1-year recurrence rate were also compared. 【Results】The operation time, postoperative hospitalization time, intraoperative bleeding volume, drainage time and the number of dissected lymph nodes of the Group B and the Group C were significantly higher than those in the Group A, and the Group C was significantly higher than the B group, the difference was significant (P<0.05). CRP and cortisol of the Group C were significantly higher than those in the Group A and the Group B (P<0.05), but there was no significant difference in WBC, FT3, FT4 and postoperative complications between the three groups(P>0.05). One year recurrence rate the Group A was significantly higher than that of the Group B and the Group C (P<0.05).【Conclusion】 The three surgical methods have their own characteristics, and the appropriate surgery should be chosen according to the conditions of patients.

Expression and Significance of Serum Vaspin and CTRP9 Levels in Patients with Polycystic Ovary Syndrome

Detection and comparison of four groups of patients with serum Vaspin, CTRP9 levels were compared between the four groups in general and biochemical indicators such as three triglycerides (TG), total cholesterol (TC), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), fasting insulin resistance index (FINS), total testosterone and insulin (HOMA-IR), and the level of steady period of 120 to 180 min glucose metabolic rate (M120-180).【Results】 The serum Vaspin level of the observation group was significantly higher than that of the control group, and the serum CTRP9 level was significantly lower than that of the control group, the difference was statistically significant (P<0.05); Pearson correlation analysis showed that the level of Vaspin was negatively correlated with HDL-C and M120-180 (P<0.05), and positively correlated with BMI, TC, LDL-C, waist circumference, FINS, TG, total testosterone, and HOMA-IR levels (P<0.05); The level of CTRP9 was positively correlated with HDL-C and M120-180 (P<0.05), and negatively correlated with BMI, TC, LDL-C, waist circumference, FINS, TG, total testosterone and HOMA-IR levels (P<0.05); Serum levels of Vaspin and CTRP9 were not significantly correlated with age (P>0.05). 【Conclusion】 Compared with the healthy population, PCOS patients have higher serum Vaspin levels and lower CTRP9 levels; The indexes of BMI, TC, HDL-C, waist circumference, FINS, TG, total testosterone, HOMA-IR, LDL-C and M120-180 were closely related to serum levels of Vaspin and CTRP9.

Analysis of the Relationship Between Surgical Treatment and Postoperative Tumor Recurrence and Postoperative Quality of Life in Patients with Primary Liver Cancer

【Objective】To analyze the relationship between surgical treatment and postoperative tumor recurrence and postoperative quality of life in patients with primary liver cancer (PLC). 【Methods】A total of 358 patients with PLC were divided into the liver resection group (n=308) and the liver transplantation group (n=50). According to different liver function, the patients were further divided into the moderate to severe cirrhosis group (n=34), mild cirrhosis group (n=230) and non-cirrhosis group (n=44). The postoperative survival rate, tumor recurrence rate and quality of life were compared between the two groups. 【Results】There were no significant differences in 1, 3 and 5-year cumulative survival rates and cumulative recurrence rate between the liver resection group and the liver transplantation group (P>0.05). The 3-year and 5-year survival rates of patients with moderate to severe liver cirrhosis were lower and the 1-year, 3-year and 5-year cumulative recurrence rates were higher than in the mild cirrhosis patients and the patients without cirrhosis (P<0.01). However, there was no significant difference between the mild cirrhosis group and the non-cirrhosis (P>0.05). The scores of physical function (PH), mental function (PS), symptoms/side effects (ST), social function (SO), self-evaluation and the total score were lower in the liver resection group than the liver transplantation group (P<0.01). Correlation analysis showed that the two operation methods and the degree of liver cirrhosis were closely related to tumor recurrence rate and quality of life. 【Conclusion】 The short-term and long-term curative effects of liver resection and liver transplantation are similar in the treatment of non-cirrhosis or mild cirrhosis. However, the tumor recurrence rate is high in patients with moderate to severe liver cirrhosis after liver resection, which will decrease the quality of life.

Expression and Role of IL-13 in Mice with Acute Lung Injury

【Objective】 To investigate the expression of IL-13 (interleukin-13) and its effect on inflammatory reaction of acute lung injury (ALI) induced by LPS in mice.【Methods】LPS was intratracheally injected into mice to induce the ALI model. The real-time PCR and ELISA were used to detect IL-13 mRNA and protein expression in lung tissue, bronchoalveolar lavage fluid (BALF) and serum. The effects of IL-13 on ALI was investigated using IL-13 neutralizing antibody. The ELISA method was used to detect the content of TNF-α and IL-1β in BALF and serum. Pretreatment of IL-13 neutralizing antibody increased the expression of TNF-α and IL-1β, and aggravated the changes in pathological damage of ALI mice (P<0.05).【Conclusion】 IL-13 expression increases in ALI induced by LPS. While inhibition of IL-13 aggravates the damage in mouse with ALI induced by LPS, which indicates that IL-13 plays a protective role in ALI endogenously.

【Objective】To study the effects of oxycodone combined with dexmedetomidine on intraoperative respiratory and circulatory functions in patients undergoing awake tracheal intubation. 【Methods】Eighty-two patients who received elective operation for difficult airway were divided into two groups with 41 cases in each group. The control group was given sufentanil combined with dexmedetomidine anesthesia, while the observation group was given oxycodone combined with dexmedetomidine anesthesia. The changes of circulatory system parameters and respiratory function indices were compared between the two groups at different time points. The sedation score (Ramsay) before intubation, comfort score after intubation, and the incidence of adverse reactions were compared between the two groups. 【Results】The levels of SBP, DBP, MAP, and HR in the observation group were significantly higher than those in the control group at 1 min and 5 min after induction (P<0.05). Analysis of I0nfluencing Factors of Left Ventricular Hypertrophy and Functional Changes in Patients with Hypertension

【Objective】To investigate the influencing factors of left ventricular hypertrophy and functional changes in patients with hypertension.【Methods】One hundred cases of patients with hypertension were selected from January 2014 to April 2016 in our hospital were selected as the research subject, according to the ambulatory arterial stiffness index (AASI) were randomly divided into observation group (AASI>0.51,n=45) and control group (AASI=0.51,n=55). Non-invasive cuff blood pressure instrument was used for automatic monitoring of patients in two groups of 24 h ambulatory blood pressure; left ventricular end diastolic diameter (LVDd), interventricular septal thickness (IVST), left ventricular posterior wall end diastolic thickness (LVDd), relative wall thickness (LVPWTD), ambulatory arterial stiffness index (AASI), left ventricular mass index (LVMI) of the two groups were measured by SIEMENS S2000 color ultrasonic diagnostic instrument; 24h mean systolic pressure (24hSBP), 24h mean diastolic pressure (24hDBP) and 24h dynamic pulse pressure (24hPP) were compared between the two groups; fasting blood glucose (FPG), three acyl glycerol (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and other biochemical indexes were compared between the two groups.【Results】24hSBP and 24hPP of the observation group were significantly higher than those of the control group, 24hDBP and 24hHR were significantly lower than those of the control group, the difference was statistically significant (both P<0.05). The difference of serum TG, TC, HDL-C levels, age, course of disease, AASI and LVMI between the observation group and the control group was statistically significant (P<0.05). The index for the two groups of patients had statistical differences in the logistics regression analysis, 24hSBP, 24hPP and AASI were independent risk factors led to the change of hypertension patients with left ventricular hypertrophy and function (P<0.05).【Conclusion】24hSBP, 24hPP and AASI are independent risk factors for left ventricular hypertrophy and functional changes in hypertensive patients. Active treatment is needed to control their blood pressure level and to improve their awareness of hypertension.

Correlation Analysis of Activity of FⅦ and Coagulation Function in Patients with Traumatic Brain Injury

【Objective】To investigate the plasma coagulation factor Ⅶ activity and its correlation with blood coagulation function in patients with acute craniocerebral trauma.【Methods】Ninety-two patients with acute craniocerebral trauma treated in our hospital from March 2014 to March 2016 were selected as the observation group, At the same time, 92 healthy volunteers in our hospital for the same period were selected as the control group, FⅦ activity value, the activated partial thromboplastin time (AFFF), platelet (PLT), two D- dimer, fibrinogen (FIB), hemoglobin (Hb), blood lactate (Lac) and other related blood coagulation index,were detected in the two groups. The international normalized ratio (INR) was calculated and intracranial pressure (ICP) was measured. The correlation between the observation group and the control group plasma FⅦ activity and its coagulation indexes were compared.【Results】FⅦ activity value of plasma of the observation group was (91.45±32.10)%, that of the control group was (97.20±16.03)%, two groups had no statistically significant difference (t=-1.537,P>0.05).INR, APTT and Lac were significantly higher than that of plasma F activity in patients with high plasma activity FⅦ. The plasma FIB level was significantly lower than that of plasma FⅦ high activity, the differences were statistically significant (P<0.05). abnormal coagulation function in patients of the observation group counted a total of 35 cases, the plasma activity of FⅦ was (86.70±28.84)%, 57 patients was accompanied with coagulation dysfunction in the observation group, The plasma activity of FⅦ was (97.83±30.16)%, there were significant differences between the two groups (t=-2.176,P<0.05).【Conclusion】Plasma activity FⅦ in patients with acute craniocerebral trauma is related to coagulation function, Plasma activities FⅦ in patients with abnormal coagulation function decreased, which is worthy of further clinical research.

Effect of Hysterectomy on Quality of Life of Patients with Refractory Functional Uterine Bleeding

【Objective】To investigate the effect of hysterectomy on quality of life of patients with refractory functional uterine bleeding. 【Methods】A total of 250 cases of patients with refractory functional uterine bleeding (observation group) who were treated by hysterectomy during January 2008 to March 2015 were selected as the observation group and 100 normal healthy women (the control group) were enrolled in the study. The Female Sexual Function Index (FSFI) was compared between the two groups. Poor quality of life in 2 years after discharge was taken as the terminal event, and the related factors influencing the quality of life were analyzed by univariate and multivariate Logistic regression analysis. 【Results】The total score of quality of life and total score of FSFI in the observation group were significantly lower than those in the control group in 3 months after operation [(121.58±19.52) vs (151.67±20.45) and (60.90±11.34) vs (75.19±12.48)](P<0.05). There were 112 cases (44.80%) marked poor quality of life and 138 cases marked good quality of life in 2 years after discharge. Multivariate Logistic regression analysis showed that age ≥50 years old was the independent risk factor for poor quality of life of patients underwent hysterectomy in 2 years after discharge (OR=1.256). 【Conclusion】Hysterectomy has adverse effect on the sexual function and quality of sexual life of patients with refractory functional uterine bleeding; age over 50 years is an independent factor affecting the quality of life after 2 years of surgery.

Effect of Magnesium Sulfate Injection under Duodenoscope on Pancreatitis and Serum Amylase Level in ERCP

【Objective】To investigate the effect of Magnesium Sulfate injection by duodenal scope on serum amylase level and the occurrence of pancreatitis in patients undergoing endoscopic retrograde cholangiopancreatography(ERCP).【Methods】A total of 82 patients undergoing ERCP in our hospital from May 2014 to May 2016 were enrolled in the study. Patients were randomly divided into magnesium sulfate group and routine group by random number table method, 41 cases in each group. All patients were treated by ERCP, endoscopic sphincteropapillotomy and endoscopic stone removal. Before intubation, magnesium sulfate was sprayed locally on duodenal papilla in magnesium sulfate group. Serum amylase, C reactive protein (CRP), Oddi sphincter pressure, success rate of intubation and incidence of complications were compared between the two groups. 【Results】The serum amylase and CRP levels of the two groups after 6 h and 24 h were significantly higher than those before the operation, and the difference was statistically significant (P<0.05); The levels of serum amylase and CRP in the Magnesium Sulfate group after 6 h and 24 h were significantly lower than those in the conventional group (P<0.05). 10 min after spraying, the SO basal pressure, SO contraction amplitude and SO contraction frequency of the two groups were significantly higher than those before spraying (P<0.05). There was no significant difference in SO interval between the Magnesium Sulfate group and the normal group after 10 min spraying (P>0.05); The SO basal pressure, SO contraction amplitude and SO contraction frequency were significantly lower than those in the conventional group (P<0.05). The successful rate of intubation in Magnesium Sulfate group was significantly higher than that in routine group, the incidence of pancreatitis and amylase was significantly lower than that of the routine group, the difference was statistically significant (P<0.05).【Conclusion】 In ERCP surgery, Magnesium Sulfate injection can relax the sphincter of Oddi by endoscopy ,which can improve the success rate of intubation, reduce the damage of pancreatic tissue, reduce the level of serum amylase and the incidence of pancreatitis.

Analysis of Sensitivity and Specificity of Clinical Features of Tracheobronchial Foreign Body Diagnosis in Children

【Objective】To investigate the clinical characteristics of tracheobronchial foreign bodies in children for their diagnostic value in sensitivity and specificity.【Methods】From September 2012 to September 2016, a total of 152 pediatric patients with suspected tracheobronchial foreign bodies were included in this study. The history of foreign body aspiration, clinical manifestation, chest X-ray examination and CT scanningwere analyzed.【Results】Tracheobronchial foreign body diagnosis was confirmed by bronchoscopy. Judged by this standard, patients with positive foreign body aspiration history,inspiratory dyspnea, diminished breath sounds on one side of the chest or asymmetric breath sounds onboth sides of the chest, X-ray and CT scan findings in the chest had a higher rate of foreign bodies present; the difference was statistically significant (P<0.05). However, differences in rates of irritating cough, fever, and lung rale were not statistically significant (P>0.05) . History of foreign body aspiration, inspiratory dyspnea, diminished breath sounds on one side of the chest or asymmetric breath sounds on two sides of the chest, chest X-ray findings, and CT scan showed a high sensitivity and positive predictive value that coincided with tracheobronchial foreign body diagnosis, but the specificity was not high. Foreign body aspiration history and chest CT scan had a high consistency with rigid bronchoscopy.【Conclusion】Inspiratory dyspnea, diminished breath sounds on one side or asymmetric breath sounds on two sides of the chest, and chest X-ray findings have some reference value for tracheobronchial foreign body diagnosis. The history of foreign body aspiration and chest CT scan can provide higher diagnostic value with more accuracy.

Levels of Serum CRP, FT3, and TNF-α in the Severity Assessment of Patients with Acute Exacerbation of Chronic Obstructive Pulmonary Disease

【Objective】The aim was to investigate the levels of serum CRP, FT3, and TNF-α level in patients with acute exacerbation of chronic obstructive pulmonary disease (AECOPD) for assessment of its severity. 【Methods】A total of 106 patients with AECOPD from October 2012 to October 2015 in our hospital were selected: 48 patients with respiratory failure were chosen to be in Group A and 58 patients without respiratory failure in Group B. Meanwhile, 30 patients without AECOPD were selected as Group C. Serum level of CRP was determined by immunoturbidimetry, FT3 was measured by chemiluminescent method, and TNF-α was measured by enzyme-linked immunosorbent assay (ELISA). The correlation between severity of disease and serum levels of CRP, FT3, and TNF-α was analyzed. 【Results】The serum levels of CRP and TNF-α showed the order of Group A >Group B > Group C (P<0.05). The serum levels of FT3, however, showed the reverse order with Group A < Group B < Group C (P<0.05). After treatment, the levels of CRP and TNF-α in patients with AECOPD were significantly lower than those before treatment (P<0.05), while the levels of FT3 were significantly higher than that before treatment (P <0.05). Before treatment, the abnormal rate of CRP was higher than the abnormal rate of WBC and the abnormal rate of neutrophils (N%) (P<0.05). 【Conclusion】The results show that serum levels of CRP, TNF-α, and FT3 are correlated with the severity of AECOPD, and could be used as reference indices for the severity of AECOPD.

Clinical Effect of Dental Implant Repair in Patients with Chronic Periodontitis

【Objective】To study the the clinical effect of dental implant repair in patients with chronic periodontitis. 【Methods】52 patients with chronic periodontitis were selected from May 2012 to June 2013 in our hospital as the observation group,50 patients with periodontal health were selected as control group. The two groups were treated with the same dental implant repair technique. The successful rate of planting in the six months, 1 years and 3 years after implantation was observed and compared between the two patients. The volume of gingival crevicular fluid (PISF), the improved plaque index (mPLI), modified sulcus bleeding index (mSBI), probing pocket depth(PPD) and the attachment loss (AL) were recorded and compared.【Results】 The success rate of the observation group in the half year and 1 year was slightly lower than that in the control group, but there was no significant difference; But retention rate of observation group was significantly lower than the control group after 3 years (P<0.05);Volume of pri-implant sulcular fluid PISF,mSBI,PPD,AL of the observation group was significantly higher than the control group; modified plaque index (mPLI) was significantly lower in the control group (P<0.05).【Conclusion】For chronic periodontitis patients, the short-term curative effect of dental implant repair technique is good, but the long-term curative effect is poor. It may be associated with chronic periodontitis and peri implant inflammation.

)], the rats were further subdivided into 4 subgroups within each group: S1, S2, S3, S4; IR1, IR2, IR3, IR4,; and RPC1, RPC2, RPC3, RPC4. The Hepatic ischemia reperfusion model in rats was established using Pringle's maneuver method. The serum levels of alanine aminotransferase (ALT) and creatinine (Cr) were examined at each time point mentioned above. After 6 hours of reperfusion, the pathological changes in the kidneys were observed. TUNEL staining was used to examine cell apoptosis, and Western blot was employed to examine protein expressions of Bcl-2 and Caspase3 in renal tissues of each group. 【Results】 Serum ALT and creatinine levels in the I/R group and RPC group increased after 30 min of ischemia-these changes were significantly higher than that in the S group (P<0.05). However, levels in RPC group were lower than those in the I/R group (P<0.05). The renal tissue structure of the rats in group S was normal. In the IR group, there were different degrees of pathological damage of renal tissue: renal tubular epithelial cell swelling, lack of structure, nucleus condensation, necrotic exfoliated epithelial cells, and inflammatory cell infiltration. The pathological damage was obviously improved following remifentanil preconditioning in RPC group. TUNEL staining revealed that there were significantly lower apoptotic cells in the S group than those in the I/R group and the RPC group (P<0.05), and that the degree in the I/R group was much higher than that in the RPC group (P<0.05). Western blot detection of apoptotic protein showed that the expression of Bcl-2 and Caspase3 in the I/R group were higher than those in the S group (P<0.05). After Remifentanil preconditioning in RPC group, the expression of Bcl-2 increased but Caspase-3 expression decreased (P<0.05).【Conclusion】 Remifentanil preconditioning has a protective effect on renal injury after hepatic ischemia reperfusion, and the protective mechanism may be involved in the regulation of renal cell apoptosis and the expression of Bcl-2 and Caspase-3.

Comparison of Efficacy and Safety of Urapidil and Nicardipine in Elderly Patients with Acute Hypertension

【Objective】To compare the clinical efficacy and safety of urapidil and nicardipine in the treatment of elderly patients with essential hypertension. 【Methods】Sixty cases of elderly hypertensive patients admitted from January, 2015 to October, 2016 were selected as the study data . According to the principle of randomization, they were divided into urapidil group and nicardipine group, 30 cases in each group. Urapidil group was given intravenous injection of 12.5 mg, then given 50~200 g/min g/min urapidil continuous intravenous infusion; nicardipine group was given intravenous injection of nicardipine 1 mg, then 10~40 Mu A, continuous intravenous drip.In combination with the changes of blood pressure in patients, the dose of drugs was optimized and adjusted. The heart rate (HR), blood pressure changes and adverse reactions were observed before and after medication in two groups.【Results】The total effective rate of urapidil group was 90% (27/30), and the total effective rate was 93.33% (28/30) in the nicardipine group, but there was no significant difference between the two groups (P>0.05). 10 min, 20 min, 30 min, 60 min and 120 min SBP of two groups of patients were significantly lower than those before treatment; 60 min, 120 min DBP and HR of two groups were significantly lower than that before treatment, the differences were statistically significant (P<0.05); The HR of urapidil group was significantly lower than that of nicardipine group at 10 min, 20 min, 30 min, 60 min and 120 min, with statistical difference (P<0.05). Two groups of patients had different degrees of adverse reactions such as dry mouth, constipation, flushing during treatment, occurrence of adverse reaction rate of 6.7% (2/30) of the urapidil group was significantly lower than that of the nicardipine group (6/30 20%), with statistically significant differences between the groups (χ

Comparison of the Effects between Laparoscopic and Open Surgery in the Treatment of Elderly Patients with Rectal Cancer

【Objective】To compare the long-term curative effect and the influence on the quality of life between laparoscopic surgery and open surgery in the treatment of elderly patients with rectal cancer. 【Methods】Ninety-six patients with rectal cancer were randomized into two group. The observation group was treated with laparoscopic radical resection of rectal cancer, while the control group was treated with open surgery. The surgical effect, 3 months postoperative quality of life score, and follow-up survival rate of patients was compared between the two groups. 【Results】The intraoperative blood loss, off-bed time, and length of hospitalization of the observation group patients were significantly better than those of control group (P<0.05). The total incidence of complications in the observation group (6.25%) was significantly lower than that in the control group (25.00%) (P<0.05). The scores of overall health, physical function, pain, mental health, and social function in the observation group patient 3 months after surgery were significantly higher than those in the control group (P<0.05). However, there was no significant difference in the 1-year and 3-year survival rates between the observation group and the control group (P>0.05). 【Conclusion】 The operation times and long-term effects of laparoscopic and open surgery in the treatment of rectal cancer are similar. However, laparoscopic surgery allows for less intraoperative blood loss, quicker postoperative recovery, and fewer complications. It can significantly improve the postoperative quality of life of patients.

【Objective】Analysis of the effect of gestational nutrition on maternal and neonatal outcomes of gestational diabetes mellitus(GDM). 【Methods】 Ninety-three cases of gestational diabetes mellitus in our hospital(January 2014 to October 2015) were selected, they were randomly divided into intervention group (Comprehensive intervention: nutrition guidance, aerobic exercise, appropriate physical exercise)and control group(Regular diet control), the research object of FBG, P2hBG, HbA1c before and after intervention were compared between the two groups. The difference of incidence of adverse pregnancy outcome and perinatal outcome were compared between the two groups. 【Results】After the intervention, intervention group of FBG, P2hBG, HbA1c were lower than the control group, and the difference had statistical significance (P<0.05). The incidence of amniotic fluid pollution in the intervention group (12 cases, 25%) was lower than that of the control group (26 cases, 57.78%), the incidence of pregnancy induced hypertension (8 cases, 16.67%) was lower than that of the control group (17 cases, 37.78%), the incidence of splits the palace production(17 cases,35.42%) was lower than that of the control group (28 cases, 62.22%), and the difference is statistically significant (P<0.05); The incidence of low birth weight infants in the intervention group (5 cases, 10.42%) was lower than that in the control group (13 cases, 28.89%), The incidence of small for gestational age infant (6 cases, 12.50%) was lower than that of the control group (15 cases, 33.33%), the incidence of neonatal hypoglycemia (2 cases, 4.17%) was lower than that of the control group (10 cases, 22.22%), the incidence of neonatal asphyxia (4 cases, 8.33%) was lower than that of the control group (11 cases, 24.44%), the incidence of the pediatric treatment (7 cases, 14.58%) was lower than that of the control group (16 cases,35.56%), and the difference is statistically significant (P<0.05). 【Conclusion】Gestational nutrition comprehensive nursing intervention can effectively reduce the incidence of GDM, adverse pregnancy outcomes and the adverse perinatal outcomes.

【Objective】To investigate the effects of surgical timing and surgical selection on clinical outcome in patients with colon cancer complicated with actual intestinal obstruction.【Methods】Sixty cases of colon cancer patients complicated by actual intestinal obstruction were collected from January to November, 2016 in our hospital for the study. All patients underwent stage I resection and anastomosis and the procedure of surgery were recorded. Among 60 cases, 20 patients received the surgery immediately after proper completion of preoperative preparation (group A), 20 received the same surgery after 8~12 hours of conservative treatment (group B), and 20 had surgery after 2~3 days of conservative treatment (group C). As to the intraoperative treatment of the bowel, 25 cases adopted traditional Dudley lavage and 35 cases were applied with modified Dudley lavage. The number of case of anastomotic fistula, abdominal infection, and death under different circumstance of bowel treatment and position of actual intestinal obstruction were analyzed and compared among the three groups. 【Results】Surgery with stage I resection and anastomosis showed good curative effect in all patients without low anastomotic fistula and abdominal infection. Patients in group A had a better clinical outcome; patients in group C were cured and discharged. There was one case of death after surgery in group B. The time of operation, hospitalization, and discharge did not show significant difference among the three groups (P>0.05). In the traditional Dudley lavage, 3 cases (3/25,12%) had anastomotic fistula, 3 cases had abdominal infection, and 1 died. However, there were no complication and death seen in the modified Dudley lavage with colon decompression. Statistically, the difference was significant in between the two methods (P<0.05). One died in 30 cases of left colon cancer complicated with actual intestinal obstruction and no death was in 30 cases of right colon cancer complicated with actual intestinal obstruction; the difference was not statistically significant (P>0.05). 【Conclusion】A certain difference exists between different times of treatment and applications of different methods in colon cancer complicated with actual intestinal obstruction. Careful selection of the surgery timing is of important.
