1448 Mechanism of LncRNA HOTAIR in Photodynamic Therapy of Colorectal Cancer
GONG Hui, LIAO Ping, YANG Qing-long, et al
DOI: 10.3969/j.issn.1671-7171.2021.10.003
【Objective】To explore the mechanism of long non-coding RNA(LncRNA)-HOX transcribed antisense RNA(HOTAIR) in photodynamic therapy (PDT) of colorectal cancer. 【Methods】The expression of lncrna hotair in colorectal cancer cells (CRCs) treated with PDT was detected by real-time fluorescence quantitative PCR (qPCR). CCK8 kit and plate clone formation experiment were used to detect the proliferation of CRCs after interfering with the expression of LncRNA HOTAIR and treated with PDT. Meanwhile, western blot assay (WB) was used to detect the expression of cell proliferation-related proteins p38 and p-p38.Then, the intersection of LncRNA HOTAIR downstream regulatory protein and PDT sequencing data was analyzed by Starbase database to obtain both the regulatory protein of LncRNA HOTAIR and the down-regulated protein after PDT treatment. Finally, after interfering with the expression of LncRNA HOTAIR and overexpressing ENPP5, the cell proliferation level was detected by CCK8 kit and plate cloning experiment, and the expression of p38 and P-38 protein were detected by WB experiment.【Results】After PDT treatment, the cell morphology of CRCs changed significantly, and the expression of LncRNA HOTAIR was significantly down regulated. Interference with LncRNA HOTAIR expression in CRCs can improve the sensitivity to PDT. LncRNA HOTAIR in CRCs can positively regulate ENPP5 expression. Interfering with the expression of LncRNA HOTAIR in CRCs can down regulate the expression of ENPP5, affect the p38 signal pathway and promote the sensitivity to PDT.【Conclusion】Interference with LncRNA HOTAIR expression increased CRCs sensitivity to PDT, and LncRNA HOTAIR positively regulated ENPP5 to affect PDT sensitivity to CRCs. It provides a new idea for improving the efficacy of PDT in colorectal cancer.

【Objective】To investigate the correlation between different alkaline phosphatase (ALP) levels and hemorrhagic transformation of cerebral infarction. 【Methods】 A total of 718 patients with cerebral infarction hospitalized in our hospital from January 2018 to January 2020 were selected. The ALP level was measured within 48 hours and the general baseline data were collected. According to the European collaborative study on acute stroke (ECASS Ⅲ), the hemorrhagic transformation (HT) of cerebral infarction was divided into hemorrhagic cerebral infarction (HI,n=33) and cerebral parenchymal hemorrhage (PH,n=123). Among them, the aggravation of neurological dysfunction caused by hemorrhagic transformation (NIHSS score increased by ≥ 4 points) was defined as symptomatic intracranial hemorrhage (sICH). The differences of ALP levels between hemorrhagic transformation group (HTG group) and non hemorrhagic transformation group (NHTG group) were compared. The correlation between different ALP levels and different types of hemorrhagic transformation was evaluated and analyzed by Quantile logistic regression analysis. 【Results】 There was no significant difference in the level of ALP between HI group, pH group and nhtg group (P>0.05). The level of ALP in the sICH group was significantly higher than that in NHTG group (P<0.05). Quantile regression analysis showed that different levels of ALP had no correlation with hi and pH, while high levels of ALP had significant correlation with sICH (P<0.05). 【Conclusion】Patients with cerebral infarction combined with high-level ALP are more prone to sICH transformation.

【Objective】To investigate the clinical efficacy of tiapride hydrochloride tablets combined with Wuling capsule in the treatment of alcoholic mental disorder. 【Methods】 A total of 86 patients with alcoholic mental disorders treated in our hospital from March 2017 to July 2020 was randomly divided into observation group (treated with tiapride hydrochloride tablets combined with Wuling capsule) and control group (treated with tiapride hydrochloride tablets), with 43 cases in each group. The efficacy, positive and Negative Syndrome Scale score (PANSS), depression (SDS), self rating Anxiety Scale score (SAS), P300 parameters and safety were compared between the two groups. 【Results】 The total effective rate of the observation group was 90.70% (39/43), which was higher than 72.09% (31/43) of the control group(χ2=4.914,P=0.027<0.05). After treatment, the scores of PANSS positive symptoms, negative symptoms, general psychopathological symptoms, SDS and SAS in the two groups were lower than those before treatment, and the observation group was lower than that in the control group (P<0.05); The latency of P300 parameters N1, N2 and P3 in the two groups were lower than those before treatment, and the amplitudes of N1, N2 and P3 were higher than those before treatment (P<0.05); The latency of P300 parameters N1, N2 and P3 in the observation group were lower than those in the control group, and the amplitudes of N1, N2 and P3 in the observation group were higher than those in the control group (P<0.05). The total incidence of adverse reactions in the observation group was 2.33% (1/43), which was lower than 9.30% (4/43) in the control group, but the difference was not statistically significant( χ2 =0.849,P=0.357>0.05). 【Conclusion】 Tiapride hydrochloride tablets combined with Wuling capsule is effective in the treatment of alcoholic mental disorder, can reduce the symptoms of anxiety and depression, and has good safety.

【Objective】To investigate the effect of paclitaxel combined with cisplatin intraperitoneal thermal perfusion chemotherapy on tumor markers and prognosis in patients with ovarian cancer.【Methods】80 women diagnosed with ovarian cancer in our hospital were randomly divided into observation group and control group, with 40 cases in each group. The control group was treated with paclitaxel combined with cisplatin intravenous drip, and the observation group was treated with paclitaxel combined with cisplatin intraperitoneal circulation thermal perfusion. The curative effects of the two groups after 4 months of treatment were compared; The levels of serum carcinoembryonic antigen (CEA), alpha fetoprotein (AFP), neuron specific enolase (NSE), carbohydrate antigen 125 (CA125) and kappa functional state score (KPS) were evaluated and compared between the two groups before and after treatment; The long-term survival and adverse reactions of the two groups were compared.【Results】the objective remission rate (ORR) in the observation group was 80.00% (32/40), which was significantly higher than 57.50% (23/40) in the control group (P<0.05). CEA, NSE and CA125 in the observation group were significantly lower than those before treatment and after treatment in the control group, and KPS scores were significantly higher than those before treatment and after treatment in the control group (P<0.05). The 1-year survival rate in the observation group was significantly higher than that in the control group (P<0.05). The total incidence of adverse reactions in the observation group was 80.00% (32/40), and there was no significant difference compared with 67.50% (27/40) in the control group (P>0.05).【Conclusion】Paclitaxel combined with cisplatin for intraperitoneal circulation thermal perfusion is more effective and safe, and has a positive impact on improving the prognosis of patients.

DOI: 10.3969/j.issn.1671-7171.2021.10.034
【Objective】To observe the expression of DKK-1, β-catenin and caspase 3 in patients with diffuse large B-cell lymphoma (DLBCL) and analyze the influencing factors. 【Methods】The patients with DLBCL who were treated in our hospital from February 2014 to August 2019were enrolled. The expressions of DKK-1, β-catenin and caspase 3 in cancer tissues and paracancerous tissues of patients with DLBCL were detected and their effects were analyzed. 【Results】The positive expression rates of DKK-1, β-catenin and caspase 3 in cancer tissues of DLBCL patients were significantly higher than those in cancer adjacent tissues. The difference was statistically significant (P<0.05). The clinical stage in Ⅲ+Ⅳ, PS score ≥2, and IDH increased. The positive expression rates of DKK-1, β-catenin and caspase 3 in DLBCL patients with clinical stage Ⅲ+Ⅳ, PS (performance status) score ≥2, high IDH (isocitrate dehydrogenase), and high risk of IPI (international prognostic index) were much higher (P<0.05). There were no significant differences in the positive expression rates of DKK-1, β-catenin and caspase 3 in patients with different ages and genders. DKK- patients with DLBCL 1. Multivariate logistica repression showed that clinical stage and IDH were the factors that affected the expression of DKK-1, β-catenin and caspase 3 expression in DLBCL patients (OR=4.865, 5.013). 【Conclusion】The positive expression rates of DKK-1, β-catenin and caspase 3 in patients with diffuse large B-cell lymphoma are high, which are affected by clinical stage and IDH.
