Correlation between Th1, Th2 Cells, Cytokines and Pulmonary Function in Children with Respiratory Syncytial Virus Pneumonia

Abstract  【Objective】To investigate the correlation between helper T cells (Th) 1, Th2 cells, cytokines and lung function in children with respiratory syncytial virus (RSV) pneumonia. 【Methods】A total 80 children with respiratory syncytial virus pneumonia treated in our hospital from November 2017 to December 2020 were selected as RSV group, and 52 infants who underwent physical examination in our hospital at the same time were selected as control group. The percentages of Th1 and Th2 cells in peripheral blood of the two groups were detected by flow cytometry, and Th1/Th2 were calculated; the cytokine interferon (IFN-γ) and interleukin (IL) - 4 levels secreted by Th1 and Th2 cells was detected by enzyme-linked immunosorbent assay (ELISA) ; the pulmonary function of the two groups was detected by pulmonary function tester, and the Th1/Th2 and serum IFN-γ,IL-4 level and pulmonary function index were compared between the two groups; Pearson was used to analyze the correlation between Th1/Th2 ,serum IFN-γ, IL-4 level and pulmonary function indexes. 【Results】 There was no significant difference in gender, age and body mass index (BMI) between the two groups (P>0.05); Percentage of Th1 cells, Th1/Th2 and IFN-γ in peripheral blood of the RSV group. The percentage of Th2 cells and IL-4 in peripheral blood were significantly higher than those in the control group (P<0.05); Minute ventilation (MV), tidal volume (VT), inspiratory time (TI), expiratory time (TE), respiratory ratio (Ti/TE) and peak expiratory flow rate (PEF) in the RSV group were significantly lower than those in the control group, and respiratory rate (RR) was significantly higher than those in the control group (P<0.05); Th1/Th2 was positively correlated with MV, VT, TE and PEF (r=0.86, 0.57, 0.34, 0.67, P<0.05), and negatively correlated with RR (r=-0.69, P<0.05); IFN-γ was positively correlated with VT, Ti/TE and PEF (r=0.47, 0.64, 0.70, P<0.05), and negatively correlated with RR (r=-0.81, P<0.05); IL-4 was negatively correlated with MV, VT, Ti/TE and PEF (r=-0.47, -0.60, -0.32, -0.63, P<0.05). 【Conclusion】 Th2 dominant response is the main manifestation of immune response in children with RSV pneumonia. Th1, Th2 cells and their secreted cytokines are significantly correlated with pulmonary function indexes, which can be used as indexes for judging the condition and evaluating the prognosis.
