摘要【目的】探讨硫氧还蛋白还原酶(thioredoxin reductase,TrxR)、血清七种自身抗体(seven autoantibodies,7-AABs),叉头框A1蛋白在早期肺癌组织中的表达水平及其意义。【方法】本院收治的100例早期肺癌患者,均在本院进行手术切除癌组织,并留取肺癌组织和距离癌组织边缘2 cm以上的癌旁组织。采用酶联免疫法检测抑癌基因产物、蛋白基因产物9.5(Protein gene product,PGP9.5)、性别决定区Y框蛋白2(The sex-determining region Y box protein 2,SOX2)、肺癌组肿瘤/睾丸抗原G抗原7(Tumor/testicular antigen G antigen 7 in lung cancer group,GAGE7)、ATP结合RNA旋酶GBU4-5、黑色素瘤抗原A1(Melanoma antigen A1,MAGEA1)、肿瘤相关基因(Tumor-associated gene,CAGE)水平,采用免疫组化学染色检测TrxR、叉头框A1蛋白水平。【结果】肺癌患者中7-AABs水平高于对照组(P<0.05),TrxR、叉头框A1蛋白表达水平高于癌旁组织;N1淋巴结转移者7-AABs、TrxR、叉头框A1蛋白表达水平高于无N1淋巴结转移者;低分化者7-AABs、TrxR、叉头框A1蛋白表达水平高于高-中分化者,且差异均具有统计学意义(P<0.05)。【结论】TrxR、7-AABs、叉头框A1蛋白在早期肺癌组织中的表达水平显著升高,且与早期肺癌有无N1淋巴结转移,分化程度有关,三者均参与了早期肺癌的发生和发展。
Abstract:【Objective】To investigate the expression and significance of thioredoxin reductase (TrxR),seven serum autoantibodies (7-AABs) and forkhead box A1 protein in patients with early lung cancer. 【Methods】A total of 100 patients with early-stage lung cancer were treated in our hospital. The cancer tissues were surgically removed,and the lung cancer tissues and adjacent tissues more than 2 cm away from the edge of the cancer tissues were retained. The tumor suppressor gene product,protein gene product 9.5 (PGP9.5),sex determining region Y box protein 2 (SOX2),tumor/testicular antigen g-antigen 7 (GAGE7),ATP binding RNA gyrase GBU4-5,melanoma antigen A1 (MAGEA1) and cancer associated gene (CAGE) were detected by enzyme-linked immunosorbent assay. TrxR and forkhead box A1 proteins were detected by immunohistochemical staining.【Results】The expression levels of 7-AABs,TrxR and forkhead box A1 protein in cancer tissues were higher than those in adjacent tissues; The expression levels of 7-AABs,TrxR and forkhead box A1 protein in patients with N1 lymph node metastasis were higher than those without N1 lymph node metastasis; The expression levels of 7-AABs,TrxR and forkhead box A1 protein in poorly differentiated patients were higher than those in highly and moderately differentiated patients,and the differences were statistically significant (P<0.05).【Conclusion】The expression of TrxR,7-AABs and forkhead box A1 protein in early lung cancer increased significantly,which was related to the presence or absence of N1 lymph node metastasis and the level of high,medium and low differentiation. All three are involved in the occurrence and development of early lung cancer.
