XIE Hong, CHEN Jian-ping, HE Jun-bin,et al
DOI: 10.3969/j.issn.1671-7171.2020.01.08
【Objective】To investigate the value of DCE-MRI semi-quantification in predicting the sensitivity of patients with lung cancer to concurrent chemoradiotherapy and relationship with tumor regression rate.【Methods】 A total of 83 lung cancer patients who underwent concurrent chemoradiotherapy in our hospital were selected.DCE-MRI scan was used to analyze the relationship of semi-quantitative parameters with tumor regression rate and chemoradiotherapy sensitivity prediction. The imaging quality of DCE-MRI images were also observed in this study. 【Results】The DCE-MRI image quality showed that there were 61 cases (74.50%), 20 cases (24.09%), 2 cases (2.41%), and 0 cases in grade 3, 2, 1 and 0, irrespectively. The sensitivity, specificity and accuracy of concurrent chemoradiotherapy sensitivity prediction by DEE-MRI were 84.6%, 83.9% and 80.4%, irrespectively, based on the ROC curve analysis (Ve=0.20). After 1 month of treatment, there were 43 cases in the sensitive group and 40 cases in the insensitive group. The TTP value in the sensitive group was lower than that in the insensitive group, while the SERmax value in the sensitive group was significantly higher than that in the insensitive group ; the difference was statistically significant (P<0.05) . The values of PET, MSI and MSD in the two groups were not significantly different (P> 0.05). The tumor area decreased gradually after radiotherapy and chemotherapy; and the tumor area at 3 months after treatment was (7.23±3.01)cm2, which was significantly lower than that before radiotherapy and chemotherapy, while the tumor regression rate presented a gradually increasing trend , the difference was statistically significant (P<0.05). Ktrans, a semi-quantitative parameter of DCE-MRI, was positively correlated with tumor regression rate in lung cancer patients before treatment (r=0.421, P=0.014), and Ve was negatively correlated with tumor regression rate (r=-0.881,P=0.000). Patients who had higher value of Ktrans before treatment showed lower value of Ve, indicating more sensitive to concurrent chemoradiotherapy.【Conclusion】 DCE-MRI semi-quantitative parameter analysis shows a high value in predicting the sensitivity of radiotherapy and chemotherapy for lung cancer. The image quality is also good. Thus it can guide the selection of chemotherapy drugs to achieve the final curative effect, which is worthy of wide application.

ZHAN Feng, LIU Qian, HE Guang-zhao,et al
DOI: 10.3969/j.issn.1671-7171.2020.01.010
【Objective】To study the relationship between the levels of CD3+CD4+T cells and CD3+CD8+T cells and the prognosis of patients with advanced non-small cell lung cancer(NSCLC).【Methods】 One hundred and eighty patients with advanced NSCLC cancer were treated in our hospital. All patients received PG chemotherapy (Pemetrexed + Nedaplatin) for 2 cycles.Peripheral venous blood samples of patients after chemotherapy were collected, and the levels of T lymphocyte subsets (CD3+CD4+, CD3+CD8+) were determined . Multivariate logistic regression analysis was used to analyze the independent influencing factors of patients' short-term prognosis and to observe the relationship between the levels of CD3+CD4+, CD3+CD8+ and patients' short-term prognosis. 【Results】 After two chemotherapy cycles of 180 patients, 43 cases (23.89%) of PR and 116 cases (64.44%) of SD were found and were included in the disease control group, while 21 cases (11.67%) of PD were included in the disease uncontrolled group; the proportion of KPS score ≥ 80, tumor diameter ≥ 5cm, low differentiation, TNM stage IV, platelet count ≥ 300×109/L in the disease control group were significantly lower than those in the disease uncontrol group (P<0.05); the levels of CD3+CD4+, CD3+CD8+ were significantly higher than those of the disease uncontrol group (P<0.05). Multivariate logistic regression analysis revealed that KPS score, tumor diameter, degree of differentiation, TNM stage and extra-platelet count were independent influencing factors for the recent prognosis of patients with advanced NSCLC (P<0.05), whereas CD3+CD4+, CD3+CD8+ levels were not independent influencing factors for the recent prognosis of patients (P>0.05).【Conclusion】The recent efficacy of chemotherapy for advanced NSCLC cancer is not significantly correlated with CD3+CD4+, CD3+CD8+ levels, suggesting that detection of peripheral blood t lymphocyte subsets could not serve as a basis for judging the efficacy of chemotherapy.

LUO Ze-bin, CHEN Zhi-an,CHEN Long-lin,et al
DOI: 10.3969/j.issn.1671-7171.2020.01.015
【Objective】To detect the expression of circRNA_0006633 in liver cancer tissues and explore its biological regulation on cell proliferation, apoptosis and cell cycle.【Methods】Real-time quantitative polymerase chain reaction (RT-qPCR) was used to analyzed the expression level of circRNA_0006633 in liver cancer tissues and adjacent normal tissues, while the relationship between circRNA_0006633's expression and clinicopathological parameters was further determined. Then biological effects of circRNA_0006633 overexpression by lentivirus-mediated-plasmids (circ_0006633-pcDNA 3.1 plasmids)on cell proliferation, apoptosis, and cycle were investigated by RT-qPCR, western blot, CCK8 and flow cytometry.【Results】Compared to the paired adjacent normal tissues, the expression level of circRNA_0006633 in liver cancer tissues was lower. Furthermore, the low expression circRNA_0006633 was negatively correlated with survival outcome. Upregulation of circRNA_0006633 effectively reduced cell proliferation, increased apoptosis and arrested cells at G2/M phages, while the epithelial-mesenchymal transition (EMT)-related proteins were regulated as well.【Conclusion】Expression of circRNA_0006633 is lower in liver cancer tissues. circRNA_0006633 can inhibit the process of epithelial-mesenchymal transition pathway activation and reduce the proliferation of liver cancer cells, which provides new ideas for the treatment of liver cancer.

Fasting blood glucose (FBG), triacylglycerol (TG), serum creatinine (SCr), blood uric acid (UA), and high-density lipoprotein (HDL-C), estimate glomerular filtration rate (eGFR), body mass index (BMI), urine protein, systolic and diastolic blood pressure were recorded. The incidence of MS in patients with different urinary protein content was observed and compared. Multivariate regression analysis was performed on the factors that may affect the urine protein content. 【Results】Among 164 patients, 62 patients in group A had MS, accounting for 37.80%; 102 patients in group B had no MS, accounting for 62.20%. The levels of systolic blood pressure , diastolic blood pressure, FBG,TG,SCr,UA,BMI and urine protein in group A were higher than those in group B, while levels of HDL-C and eGFR in group A were lower than those in group B (P<0.05). With increasing urinary protein content, the eGFR of patients in group Ⅰ,Ⅱ and Ⅲ gradually decreased. The eGFR of group Ⅰ was significantly higher than that of group Ⅱ and group Ⅲ (P<0.05). Systolic blood pressure , diastolic blood pressure, FBG , TG、HDL-C、SCr、BMI and MS incidence rate in group Ⅰ were lower than those in group Ⅱ and Ⅲ (P<0.05). Increased waist-to-hip ratio was the only independent risk factor for the occurrence of moderate proteinuria, while mildly elevated blood glucose, increased TG/HDL-C ratio, advanced age and increased waist-to-hip ratio were independent risk factors for the development of massive proteinuria.【Conclusion】Patients with chronic kidney disease patients have a high rate of metabolic syndrome. Fasting blood glucose, increased waist-to-hip ratio and dyslipidemia are independent risk factors for the development of massive proteinuria in patients with chronic kidney disease. Therefore, early intervention of MS can effectively delay the progression of chronic kidney disease.

DOI: 10.3969/j.issn.1671-7171.2020.01.025
【Objective】To investigate the effect of lower respiratory tract infection on the relative expression of beta-catenin in patients with chronic obstructive pulmonary disease (COPD).【Methods】A total of 94 patients with chronic obstructive pulmonary disease diagnosed and treated in the Department of Respiratory Medicine of the People's Hospital of Wuhan University were selected and divided into COPDⅠ(n=29) group, COPDⅡ(n=35) group, and COPDⅢ(n=30) group according to the pulmonary function classification of patients. According to whether the patients had lower respiratory tract infection, they were further divided into the infected group (n=56) and uninfected (n=38) group. The influence of lung function and respiratory tract infection on the relative expression of β-catenin was detected and analyzed. The relative expression of β-catenin in predicting the value of lower respiratory tract infection in patients with COPD was analyzed by ROC curve. 【Results】The FVC (forced vital capacity), FEV1 (forced expiratory volume in the first second), FEV1/FVC and β-catenin expression were statistically significant in the lung function of the patients with different COPD condition (P<0.05). With the aggravation of the COPD patients, the FVC, FEV1, FEV1/FVC decreased significantly (P<0.05), and the relative expression of β-catenin increased significantly (P<0.05). The FVC, FEV1 and FEV1/FVC in the infected group were significantly lower than those in the uninfected group (P<0.05), while the relative expression of β-catenin in the infected group was significantly higher than that in the uninfected group (P<0.05). The relative expression of β-catenin was negatively correlated with FVC, FEV1 and FEV1/FVC (P<0.05). The ROC curve was used to analyze the predictive value of the relative expression of beta -catenin in respiratory tract infection in patients with chronic obstructive pulmonary disease (COPD). The area under the ROC curve of the relative expression of β-catenin for the diagnosis of lower respiratory tract infection in COPD patients was 0.875.【Conclusion】There is a significant correlation between the abnormal expression of Wnt/beta-catenin signaling pathway and pulmonary function in COPD patients. The change of relative expression of beta-catenin in COPD patients caused by lower respiratory tract infection suggest it has a certain diagnostic value for COPD patients.

LIANG Gang, WANG Zhi-xiang, HE Meng-guo,et al
DOI: 10.3969/j.issn.1671-7171.2020.01.030
【Objective】To investigate the expression of Slit2 and Robo1 proteins in pancreatic cancer and their relationship with microvessel density (MVD).【Methods】 Immunohistochemical staining SP method was used to detect the expression of Slit2 and Robo1 proteins in 30 cases of pancreatic cancer tissues and 30 cases of adjacent tissues. The expression changes of Slit2 protein and Robo1 protein under different clinicopathological conditions were observed. The microvessel density (MVD) marker CD105 was detected to study the relationship between Slit2 /Robo1 protein expression and MVD in pancreatic cancer. 【Results】The positive rates of Slit2 and Robo1 were 60.00% and 70.00% in pancreatic cancer tissues, respectively; and the positive rates in the adjacent tissues were 33.33% and 40.00%, respectively. The differences between the two types of tissues were statistically significant (P<0.05). There was a positive correlation between the positive expression of Slit2 protein and Robo1 protein (r=0.818, P< 0.05). The expression of MVD in pancreatic cancer tissues was higher than that in the adjacent tissues (t=9.475, P<0.05). The MVD in pancreatic cancer tissues with positive expression of Slit2 and Robo1 was much higher than that in pancreatic cancer tissues with negative expression of Slit2 and Robot1 (t=2.048, P<0.05,t=2.897, P<0.05). Expression of Slit2, Robo1 and MVD were significantly associated with tumor size, pathological type, clinical stage, and lymph node metastasis of pancreatic cancer (P<0.05).【Conclusion】 The increased expression of Slit2 protein and Robo1 protein in pancreatic cancer tissue is closely related to the growth, invasion and metastasis of pancreatic cancer. The mechanism may be that Slit2/Robo1 signaling pathway promotes the occurrence and development of pancreatic cancer by inducing tumor angiogenesis. The expression intensity of Slit2 /Robo1 and the enumerations of MVD can be used to predict the progression and prognosis of pancreatic cancer.

DENG Feng, GONG Xiao-wei, YANG Jing
DOI: 10.3969/j.issn.1671-7171.2020.01.031
【Objective】To explore the stimulator of interferon genes (STING) signaling in the development and prognosis of gastric cancer.【Methods】A total of 210 patients with gastric cancer who underwent surgical resection in our hospital were selected for the study. Immunohistochemistry was used to detect the STING expression in gastric cancer tissue and adjacent tissues of surgical resection. The follow-up data of patients were included. Then Kaplan-Meier and Log-rank tests were applied for analyzing the relationships between the prognosis and STING expression. Cox regression model was used to analyze the influencing factors of tumor-free survival time as well. 【Results】The positive rate of STING in gastric cancer tissues was higher than that in adjacent tissues (P<0.05). According to TNM staging, among those 210 patients, 55 cases were categorized in stage Ⅰ, 33 cases were in stage Ⅱ, 122 cases were in stage Ⅲ. Follow-up was completed in 195 patients accounting for 92.9% (195/210). Only 15 cases were lost to follow-up. The mean follow-up time was (39.8±10.5) months. The positive expression of STING were shown in 77 cases (36.7%, 77/210). Recurrence and metastasis occurred in 142 patients. Univariate analysis showed that larger tumors, higher staging (T stage, N stage and TNM stage) and positive STING expression increased the risk of recurrence in patients with gastric cancer treated by surgery [HR (95%CI):1.93 (1.42~2.84), 3.95 (2.61~5.84), 3.25 (2.17~4.86), 4.91 (3.63~7.52), 2.15 (1.33~3.53)]. Compared to patients with negative STING expression, Cox regression analysis showed that patients with positive STING expression had an increased risk of gastric cancer recurrence [HR: 1.98, 95% confidence interval (CI): 1.23~3.32].【Conclusion】 STING is proposed as a novel independent prognostic factor for gastric cancer. Patients with positive expression of STING have a higher risk of postoperative recurrence.

DOI: 10.3969/j.issn.1671-7171.2020.01.032
【Objective】To investigate the relationship between magnetic resonance imaging and clinical stage in patients with hepatic alveolar hydatidosis (hepatic echinococcosis), in order to provide a theoretical basis for early diagnosis, treatment and improvement of prognosis of hepatic alveolar echinococcosis.【Methods】The imaging data of 74 patients with hepatic alveolar echinococcosis treated in the Department of Gastroenterology of our hospital from January 2010 to January 2018 were analyzed retrospectively. All patients received MRI scanning, and the MRI characteristics of the patients were analyzed and compared. According to Kodama classification principle, the lesions of hepatic alveolar echinococcus were divided into Kodama Ⅰ~Ⅴ types. The correlation between Kodama classification and the largest diameter of hepatic alveolar echinococcus was observed by Spearman correlation analysis model, and the correlation between different stages of PNM and the largest diameter of hepatic alveolar echinococcus was observed. 【Results】A total of 98 lesions with a diameter of 1.25~26.52 cm were found in 74 patients. 34 lesions in P1, 7 in P2, 53 in P3, 4 in P4, 82 in M0, 16 in M1, 53 in N0 and 45 in N1; Kodama Ⅲ and Ⅳ are the most common types in the lesions of echinococcosis, while Kodama Ⅰ, Ⅱ and Ⅴ were relatively few. Spearman's correlation analysis showed that there was a significant positive correlation between the maximum diameter of hepatic alveolar echinococcus and P stage (r=0.518, P<0.05); there was a significant positive correlation between the maximum diameter of hepatic alveolar hydatid focus and N stage (r=0.755, P<0.05); there was no correlation between the maximum diameter of hepatic alveolar hydatid focus and M stage (r=0.216,P>0.05); there was no correlation between the Kodama classification and the maximum diameter of hepatic alveolar hydatid focus (r=0.214,P>0.05).【Conclusion】Kodama Ⅲ and Ⅳ were the most common types in Kodama, P1, P3, M0 and N0 are the most common types in PNM, and there was a significant positive correlation between the maximum diameter of hepatic Echinococcus and P and N stages.
