Abstract:【Objective】 To investigate the value of acetylcholinesterase (AChE) staining and Calretinin immunohistochemical staining in the preoperative pathological diagnosis of hirschsprung's disease (HD). 【Methods】 The general clinical data of 89 children with chronic constipation,abdominal distension,or other suspected congenital megacolon (but not obvious symptoms) admitted to our hospital from April to December 2021 were retrospectively analyzed,including 39 children with HD (HD group) and 50 children without HD (non HD group). The characteristics of Calretinin immunohistochemical staining and AChE staining in HD and non HD children were analyzed,and their accuracy and sensitivity in the diagnosis of HD were analyzed. 【Results】 In the HD group,AChE staining was positive in 38 cases and negative in 1 case,while in the non HD group,AChE staining was positive in 1 case and negative in 49 cases. In the HD group,the Caletinin immunohistochemical staining was negative,while in the non HD group,the Caletinin immunohistochemical staining was positive in 49 cases and negative in 1 case. The specificity and sensitivity of AChE staining in the diagnosis of HD were 97.4% and 98.0%,respectively. The sensitivity and specificity of Caletinin immunohistochemical staining in the diagnosis of HD were 100% and 98%,respectively. 【Conclusion】 The combination of AChE staining and Calretinin immunohistochemical staining in colonic mucosa biopsy specimens is a reliable method for preoperative diagnosis of HD,and is worthy of clinical application.
