Abstract:【Objective】To discuss GADD45α regulatory effect of liver regeneration in cirrhotic rats after combined associating liver partition and portal vein ligation for staged hepatectomy(ALPPS). 【Methods】Ninety male SD cirrhotic rats fed with high-fat diet and induced by carbon tetrachloride were randomly divided into three groups:ALPPS group, surgical control group and normal control group, with 30 rats in each group. The ALPPS group was treated with ALPPS combined with hepatic septation and portal vein ligation. The operation control group only opened and closed the abdominal and perihepatic ligaments, and the normal control group was fed naturally. Ten rats in each group were killed at 1 day, 7 days and 14 days after operation. The expression of rat GADD45α gene was analyzed by Real time PCR, and the expression of rat GADD45α protein was detected by Western blotting. The liver function and liver regeneration rate of the three groups at different time points were analyzed and compared.【Results】On the 14th day after operation, the levels of serum ALT and AST in the ALPPS group were lower than those on the 1st and 7th day after operation, and also lower than those in the normal control group and the operation control group (P<0.05); The expression of GADD45α mRNA and GADD45α protein in the ALPPS group was significantly higher than that in the normal control group and the operation control group (P<0.05); The liver regeneration rate of rats in the ALPPS group was higher than that in 1 day and 7 days after operation, and higher than that in the normal control group and the operation control group (P<0.05). 【Conclusion】GADD45α has regulatory effect on liver regeneration after combined liver septation and portal vein ligation in cirrhotic rats, and can improve the rate of liver regeneration in rats.
