Abstract:【Objective】To investigate the expression and significance of microRNA-17 (miR-17) and autophagy gene-12 (ATG12) in bone marrow mono-nuclear cells (BMMNC) of adult acute lymphoblastic leukemia (ALL). 【Methods】 A total of 83 adult bone marrow specimens of ALL patients admitted to our hospital were collected as the observation group. According to their disease conditions,they were divided into the newly diagnosed group of 30 cases,the remission group of 32 cases, and the refractory group of 21 cases. A total of 30 bone marrow specimens from patients with normal bone marrow and non-hematopoietic system diseases who were treated in our hospital during the same period,which matched the age and sex of the observation group,were selected as the control group. The relative expressions of miR-17 and ATG12 in BMMNC were detected and the relationship of relative expressions of miR-17 and ATG12 were analyzed . 【Results】The relative expressions of miR-17 and ATG12 in the observation group were higher than those in the control group (P<0.05).The relative expressions of miR-17 and ATG12 in the newly diagnosed group and the refractory relapse group were significantly higher than those in the control group (P<0.05),while the relative expressions of miR-17 and ATG12 in the remission group were significantly lower than those in the newly diagnosed group and the refractory relapse group (P<0.05). The relative expressions levels of miR-17 and ATG12 in the remission group and the control group showed no statistically significant difference (P>0.05),while the relative expressions levels of miR-17 and ATG12 in the refractory and relapsed group showed no statistically significant difference from the newly diagnosed group (P>0.05). The relative expression level of miR-17 in the newly diagnosed patients with WBC ≥ 50×109/L was significantly higher than that in the patients with WBC< 50×109/L (P<0.05),while the relative expression level of ATG12 was significantly lower than that in the patients with WBC< 50×109/L (P< 0.05). The relative expression level of miR-17 and ATG12 in the patients with B-ALL was significantly higher than that in the patients with T-ALL (P<0.05).The relative expression levels of miR-17 and ATG12 were independent of gender,age,chromosome abnormality and prednisone induction sensitivity (P>0.05). Pearson correlation analysis showed that the relative expression levels of miR-17 and ATG12 were positively correlated (r=0.414,P<0.05) 【Conclusion】Both miR-17 and ATG12 are highly expressed and they are positively correlated in BMMNC of adult ALL patients. The autophagy activity of ATG12 was enhanced,suggesting that changes in the expression of miR-17 and ATG12 may be related to the occurrence,development and recurrence of ALL,whose specific mechanism of action remains to be further studied.
