5-羟色胺转运体基因多态性与青少年自伤、自杀行为的相关性

摘要 【目的】探讨5-羟色胺转运体(5-HTT)基因多态性与青少年自伤、自杀行为的相关性。【方法】选取2020年11月至2021年10月本地区226例青少年为研究对象,根据研究对象是否发生自伤、自杀行为将其分为发生组和未发生组。采集外周静脉血,提取血液基因组DNA,采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)测定5-HTT基因多态性,并确定其基因型分布。Hardy-weinberg遗传平衡定律验证研究对象基因型频率是否具有代表性。比较发生组和未发生组的5-HTT基因型分布及等位基因频率,采用Logistic回归分析影响青少年自伤、自杀行为发生的危险因素。【结果】本研究调查对象5-HTT基因型频率实际值与理论值经Hardy-Weinberg遗传平衡检验,差异均无统计学意义(P>0.05)。发生组单亲家庭、醉酒、离家出走、孤独、失恋、学习压力占比高于未发生组(P<0.05)。发生组SS基因型频率、S等位基因频率高于未发生组(P<0.05)。Logistic回归分析结果显示,醉酒、失恋、学习压力大、5-HTT基因型为SS/SL、5-HTT等位基因为S均为影响青少年自伤、自杀行为发生的危险因素(P<0.05)。【结论】5-HTT基因多态性与青少年自伤、自杀行为有关,5-HTT基因为SS型可能是青少年自伤、自杀的易感基因,S等位基因可能是致使青少年自伤、自杀的危险等位基因。

Abstract:【Objective】To investigate the association between the polymorphism of serotonin transporter (5-HTT) gene and adolescent self-injury and suicide behavior. 【Methods】 A total of 226 adolescents in the region from November 2020 to October 2021 were selected as the research subjects. According to whether the subjects had self-injury or suicide behavior,they were divided into the occurrence group and the non-occurrence group. Peripheral venous blood was collected to extract blood genomic DNA,and the 5-HTT gene polymorphism was determined using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).The Hardy-weinberg genetic equilibrium law was used to verify whether the genotype frequency of the subjects was representative. The 5-HTT genotype distribution and allele frequency were compared between the occurrence group and the non-occurrence group,and the risk factors affecting adolescent self-injury and suicide behavior were analyzed using logistic regression.【Results】 The genotype frequency of 5-HTT gene in the subjects of this study was tested by Hardy-weinberg genetic equilibrium,and the difference was not statistically significant(P>0.05). The occurrence group had a higher proportion of single-parent families,drunkenness,running away from home,loneliness,lovelorn,and learning pressure than the non-occurrence group(P<0.05). The frequency of SS genotype and S allele in the occurrence group was higher than that in the non-occurrence group(P<0.05). The logistic regression analysis showed that drunkenness,lovelorn,learning pressure,5-HTT genotype as SS/SL,and 5-HTT allele as S were all risk factors affecting the occurrence of adolescent self-injury and suicide behavior(P<0.05). 【Conclusions】The polymorphism of 5-HTT gene is associated with adolescent self-injury and suicide behavior.The SS genotype of 5-HTT gene may be a susceptibility gene for adolescent self-injury and suicide,and the S allele may be a risk allele for adolescent self-injury and suicide.
