Abstract:【Objective】To investigate the effect of cytokine signal transduction inhibitor 6 (SOCS6) on the injury of prostatitis epithelial cells. 【Methods】Normal prostate tissue cells were divided into the normal control group,the model group and the SOCS6 model group. The cells were transfected with PCDNA3.1-SOCS6 and stimulated with lentivirus infection. The expression level of SOCS6 mRNA in the cells was detected by qRT-PCR.Expression of SOCS6 protein,cysteine-containing caspase-3 (C-caspase-3) and cysteine-containing caspase-9(C-caspase-9) was detected by Western blot. MTT was used to detect cell proliferation and flow cytometry was used to detect cell apoptosis.【Results】The expression levels of SOCS6 protein and SOCS6 mRNA in the SOCS6 up-regulated group were significantly higher than those in the model group and the normal control group,and the differences were statistically significant (all P<0.05). The survival rate of cells in the SOCS6 up-regulation group was higher than that in the model group,and the apoptosis rate was lower than that in the model group (P<0.05). Compared to the normal control group,the survival rate and apoptosis rate of SOCS6 up-regulated group were not significantly different (P>0.05). The results of Western blot assay showed that the protein levels of C-caspase-3 and C-caspase-9 in the SOCS6 up-regulated group were lower than those in the model group and higher than those in the normal control group,and the differences were statistically significant (all P<0.05).【Conclusion】The low expression of SOCS6 protein in prostatitis tissue can increase the proliferation and apoptosis of epithelial cells,which is of great value for clinical diagnosis and treatment and prognosis prediction.
