Abstract:【Objective】 To investigate the effect of estrogen on the expression of epithelial mesenchymal transition markers of E-cadherin and vimentin in SPC-A-1 cell line.【Methods】 SPC-A-1 cell line was stimulated with different concentrations of estrogen; the expression of EMT markers was detected by Q-RT-PCR and Western blot; the effect of different concentrations of estrogen on cell migration was detected by Transwell test. The difference between the groups was calculated.【Results】 When the estrogen concentration was 1×10-9 mol/L、1×10-8 mol/L、1×10-7 mol/L, the Vimentin mRNA expression was (2.21±0.53), (4.81±0.66), (2.27±0.51), which was significantly higher than that of the blank control group (P<0.05). The mRNA expression of E-cadherin was (0.67±0.18), (0.42±0.14), (0.61±0.11), which was significantly lower than that of the blank control group (P<0.05). The number of cell migration was (23.65±3.25), (37.88±5.66) and (22.17±2.98) respectively (P<0.05). When the estrogen concentration was 1×10-8 mol/L, the expression of vimentin was the highest, the expression of E-cadherin was the lowest, and the number of cell migration was the highest.【Conclusion】 Appropriate concentration of estrogen can promote the expression of vimentin, inhibit the expression of E-cadherin, and improve the ability of cell migration. When the concentration of estrogen is 1×10-8 mol/L, the effect of promoting vimentin expression, inhibiting E-cadherin expression and promoting cell migration is most significant. Estrogen can promote the EMT process of SPC-A-1 cell line.
