miR-21-5p对多发性骨髓瘤细胞增殖、迁移、侵袭及STAT3信号通路的影响

摘要 【目的】探讨微小RNA(miR)-21-5p对多发性骨髓瘤(MM)细胞增殖、迁移、侵袭及转录激活因子3(STAT3)信号通路的影响。【方法】体外培养人MM RPMI-8226细胞,分为对照组(不进行转染)、siRNA NC组(转染nonsense siRNA)和miR-21-5p siRNA组(转染miR-21-5p siRNA)。实时荧光定量PCR(qRT-PCR)法检测人MM RPMI-8226细胞中miR-21-5p与STAT3表达;CCK-8法检测RPMI-8226细胞增殖;Transwell法检测RPMI-8226细胞迁移、侵袭;蛋白印迹(WB)法检测RPMI-8226细胞中p-STAT3、STAT3、Ki-67、基质金属蛋白酶-9(MMP-9)蛋白表达。【结果】与对照组、siRNA NC组相比,miR-21-5p siRNA组RPMI-8226细胞中miR-21-5p、STAT3 mRNA表达水平、细胞OD值、迁移细胞数、侵袭细胞数、p-STAT3/STAT3、Ki-67、MMP-9蛋白表达水平均显著降低(P<0.05)。【结论】下调miR-21-5p表达可以抑制MM RPMI-8226细胞增殖、迁移及侵袭,并使STAT3信号通路活化受阻。

Abstract:【Objective】 To investigate the effects of microRNA (miR)-21-5p on the proliferation, migration, invasion, signal transduction and activator of transcription 3 (STAT3) signal pathway of multiple myeloma (MM). 【Methods】 Human MM RPMI-8226 cells were cultured in vitro and divided into control group (without transfection), siRNA NC group (transfected with nonsense siRNA) and miR-21-5p siRNA group (transfected with miR-21-5p siRNA). The expressions of miR-21-5p and STAT3 in RPMI-8226 cells were detected by real-time fluorescence quantitative PCR (qRT-PCR); The proliferation of RPMI-8226 cells was detected by CCK-8 method; The migration and invasion of RPMI-8226 cells were detected by Transwell method; Western blot (WB) was used to detect the expression of p-STAT3, STAT3, Ki-67 and matrix metalloproteinase-9 (MMP-9) in RPMI-8226 cells.【Results】 Compared with the control group and the siRNA NC group, the expression levels of miR-21-5p, STAT3 mRNA, cell OD value, number of migrating cells, number of invasive cells, p-STAT3/STAT3, Ki-67 and MMP-9 protein in RPMI-8226 cells in the miR-21-5p siRNA group were significantly lower (P<0.05). 【Conclusion】 Down-regulating the expression of miR-21-5p can inhibit the proliferation, migration and invasion of MM RPMI-8226 cells, and block the activation of STAT3 signaling pathway.
