Relationship between Aminoacyl-tRNA Synthetases and Survival Probability of Lung Adenocarcinoma
LI Yan, XIONG Yan-lu, SI Yue,et al
DOI: 10.3969/j.issn.1671-7171.2019.10.001
【
Objective
】To explore the association between aminoacyl-tRNA synthetases and survival probability of lung adenocarcinoma via integrated analysis. 【
Methods
】We first obtained the gene expression data and clinical records of lung adenocarcinoma from the TCGA platform. Then we used GSEA analysis to explore the overall expression status of the aminoacyl-tRNA synthetase family in lung adenocarcinoma. Finally, we analyzed the relationship of the metabolic enzymes with total survival and progression-free survival of lung adenocarcinoma. 【
Results
】We found that the aminoacyl-tRNA synthetase family was highly expressed in lung adenocarcinoma. Survival analysis revealed that seven out of these enzymes were associated with overall survival of lung adenocarcinoma, and four enzymes were associated with progression-free survival. Patients with high expression of these enzymes showed worse prognosis. 【
Conclusion
】Aminoacyl -tRNA synthetases are highly expressed in lung adenocarcinoma and are closely related to survival probability. This provides a reference for understanding the metabolic characteristics and prevention methods of lung adenocarcinoma.

LI Qiao-rong, LONG Xiang-dang, YAO Sui,et al
DOI: 10.3969/j.issn.1671-7171.2019.10.003
【
Objective
】To investigate the clinical features of single umbilical artery (SUA) with cardiovascular malformations. 【
Methods
】A total of 112 fetuses with SUA were retrospectively analyzed. Clinical data and ultrasound images were collected to investigate complication with cardiovascular malformations and pregnancy outcomes. 【
Results
】Among the 112 SUA patients, simple SUA occurred in 80 cases(71.4%) and SUA complicated with other structural malformations were seen in 32 cases (28.6%),of which 25 cases(25/32,22.3%) were combined with cardiovascular malformations including 18 cases of severe complex malformations. No abnormal chromosome findings were shown in all 25 cases of SUA with cardiovascular malformations. However,20 cases were induced labor and 5 live births (2 cases of ventricular septal defect and 3 cases of ventricular septal defect with left superior vena cava were diagnosed by prenatal and postnatal ultrasound follow-up). The cardiovascular malformations in the top three places were ventricular septal defect (17/25, 68%), persistent left superior vena cava (6/25, 24%) and persistent arterial trunk (5/25, 20%). 【
Conclusions
】The most common malformations in SUA is cardiovascular malformation, which is so complicated types and poor prognosis. The diagnosis of fetal structural abnormalities during pregnancy, especially in the second trimester, is important for early intervention and reduction of birth defects.

HU Yuan,LIU Qian-jun,XIAO Li-miao,et al
DOI: 10.3969/j.issn.1671-7171.2019.10.010
【
Objective
】To investigate the value of two-dimensional speckle tracking imaging in the assessment of left ventricular systolic function in children with left ventricular noncompaction (LVNC). 【
Methods
】 A total of 27 patients with clinically diagnosed LVNC whose left ventricular systolic function were improved after standard anti-heart failure treatment were collected as the observation group. In addition, 27 healthy children were selected as the control group. The routine echocardiographic function parameters was examined firstly, then the left ventricular global longitudinal strain (GLS), global circumferential strain (GCS) and segmental longitudinal strain (LS) and circumferential strain (CS) were evaluated by two-dimensional speckle tracking imaging. The above parameters between LVNC children (pre-treatment and post-treatment) and the control group were compared. 【
Results
】After treatment, LVESV (left ventricular end-systolic volume) and LVEDD (Left ventricular end-diastolic diameter) in LVNC children were lower than those before treatment, while LVEF (left ventricular ejection fraction) was higher than that before treatment in LVNC children (all P<0.05). There were no significant differences in left ventricular end-diastolic volume (LVEDV) and LVEDD between the observation group post-treatment and the control group (P>0.05), while LVEF was significantly lower than the control group. After treatment, the left ventricular GLS, LS, GCS and CS of LVNC children were higher than those before treatment (all P<0.05). Compared to the control group, GLS, GCS, CS and 4 apical segments of LS decreased in post-treatment LVNC children (P<0.05),however, there was no significant difference of LS for the remaining segments between the control group and LVNC children post-treatment (P>0.05). In the post-treatment group of LVNC children, 25 patients had LVEF≥50% and the absolute value of LVEF increased by at least ≥10% compared with baseline. There were only 2 patients with LVNC had LVEF<50% and the absolute value of LVEF increased by <10% compared with baseline. During the follow-up period, 25 patients in the observation group kept LVEF ≥ 50% and the absolute value of LVEF increased by at least ≥ 10% compared with baseline; those two LVNC children with LVEF < 50% had no significant changes of the absolute value of LVEF compared with that before treatment. According to the modified ROSS classification, the cardiac function of the 27 HVNC children pre-treatment were grade Ⅳ. After treatment, the cardiac function of 19 children were grade II, 6 children were grade I, and 2 children were grade Ⅳ.【
Conclusion
】Left ventricular systolic function of children with LVNC get improved after standard anti-heart failure treatment, even the contraction of left ventricle segment does not return to normal level. Two-dimensional speckle tracking imaging can evaluate the left ventricular global and regional myocardial function pre- and post-treatment in children with LVNC, which provides useful reference for diagnosis and treatment of HVNC.

Effect of microRNA-132 on Gene Expression Related to Retinal Neovascularization (RNV) in Human Retinal Microvascular Endothelial Cells (HRMECs) under Hypoxia/Reoxygenation (H/R)
ZHANG Li-xin
DOI: 10.3969/j.issn.1671-7171.2019.10.013
【
Objective
】To explore the effect of microRNA-132 on gene expression related to retinal neovascularization (RNV) in human retinal microvascular endothelial cells (HRMECs) under hypoxia/reoxygenation (H/R). 【
Methods
】HRMECs cell lines were cultured under hypoxia for 6 hours and then under normal oxygen for 6 hours as H/R group; anti-mir-132 was added as H/R + anti-mir-132 group under H/R condition; HRMECs were cultured under normal oxygen condition as negative control group. Total cell RNA was extracted and the changes of gene expression patterns were detected on the RNA microarray. Pathway analysis was performed using Rosetta Resolver 7.2 software package. Finally, candidate genes related to RNV were identified with Pubmed database. 【
Results
】Compared with the negative control group, there were significant changes in cell signals such as cell adhesion, adipocytokine and extracellular matrix receptor interaction in the H/R group (P<0.05). By inhibiting the expression of microRNA-132, we found that microRNA-132 participated in the regulation of various signal molecules in H/R cells. Three RNV related genes were screened out based on the analysis of Pubmed database: THBS1, IL-6 and CXCL8. 【
Conclusion

DOI: 10.3969/j.issn.1671-7171.2019.10.016
【
Objective
】To analyze the clinical efficacy and postoperative recurrent risk of minimally invasive thoracoscopic surgery in the treatment of bilateral pulmonary bullae with spontaneous pneumothorax. 【
Methods
】A total of 94 patients with bilateral pulmonary bullae and spontaneous pneumothorax, who were treated in our hospital from June 2016 to May 2017 were retrospectively analyzed. According to different surgical methods, patients were randomly divided into the observation group (n=50 cases) and the control group (n=44 cases). The control group received routine thoracotomy and the observation group received minimally invasive thoracoscopic surgery. The clinical efficacy and operative indicators of the two groups were compared. The postoperative complications and related risk factors of postoperative recurrence were analyzed between the two groups. 【
Results
】The total effective rate of the observation group was 94.00%, which was significantly higher than that of the control group 75.00% (P<0.05). The intraoperative blood loss of the observation group was less than that of the control group. The operation time, chest tube drainage time and hospitalization time in the observation group were shorter than those in the control group. The differences between the two groups were statistically significant (P<0.05). The incidence of complications in the control group was 18.19%, which was significantly higher than that of the observation group 4.00% (P<0.05). However, there was no significant difference in recurrence rates between the two groups (P>0.05). The incidence of recurrent pneumothorax and chest tube indwelling time more than 8 day in control group was significant higher than that in observation group (P<0.05). 【
Conclusion
】Minimally invasive thoracoscopic surgery improves the clinical efficacy of bilateral pulmonary bullae combined with spontaneous pneumothorax and reduces the incidence of postoperative complications. Our results suggests that recurrent pneumothorax and chest tube indwelling time >8d may be the main risk factors for postoperative recurrence.

ZHANG Qing, ZHANG Yu-jun
DOI: 10.3969/j.issn.1671-7171.2019.10.019
【
Objective
】To investigate the correlation between the imaging characteristics of thyroid micro carcinoma (TMC) and the levels of serum TSH and VEGF. 【
Methods
】From November 2015 to October 2018, 60 patients with TMC in our hospital were selected. Bravfv600e mutation was detected to be positive in all patients by thyroid papillary carcinoma gene test. The bilateral thyroid and cervical lymph nodes were scanned by Siemens S2000 ultrasound imager. The levels of VEGF and TSH in serum were measured by immunohistochemistry, and the bilateral thyroid and cervical lymph nodes in patients with different levels of VEGF and TSH were compared.【
Results
】In this study, the mean TSH level of 60 patients was (4.23 ± 1.58) mu / L, which was divided into high TSH group (≥4.23 mu /L) and low TSH group (<4.23 mu/L).The results showed that there were significant differences in the maximum diameter, aspect ratio (A / T), boundary definition, membrane invasion and microcalcification between the two groups (P<0.05). According to the level of VEGF, they were divided into high VEGF group (≥ 27.34 U / ml) and low VEGF group (< 27.34 U / ml). The results showed that there were statistical differences in the maximum diameter of tumor, boundary clarity, membrane invasion and microcalcification between the two groups (P<0.05). The Pearson correlation of serum TSH and VEGF in TMC patients showed that the correlation coefficient of serum TSH and VEGF was 0.336, which was statistically significant (P<0.05).【
Conclusion
】The correlation between thyroid imaging characteristics of TMC and serum TSH, VEGF levels is mainly due to the common features of tumor benign and malignant judgment, so the combined detection of the two may improve the sensitivity and specificity of diagnosis.

DOI: 10.3969/j.issn.1671-7171.2019.10.020
【
Objective
】To investigate the expression of CAP2 gene in HCC and its prognostic value.【
Methods
】The HCC related gene CAP2 was screened in Oncome database, and then the secondary analysis was carried out in Oncome and CCLE database, and the prognosis analysis was carried out in gepia database. 【
Results
】Based on Oncome database, the analysis of HCC miRNA microarray showed that CAP2 was up-regulated most significantly in HCC gene expression profile. A total of 412 different CAP2 studies were included in oncoming database, among which 33 were statistically significant, 14 were highly expressed, and 19 were low expressed. A total of 8 studies found that CAP2 was highly expressed in HCC (P<0.05), a total of 646 samples; then CCLE data was used to verify the expression level of CAP2 in HCC cells, and the results showed that CAP2 was highly expressed, consistent with oncomine. There was correlation between CAP2 and the disease-free survival rate of HCC patients (P<0.05), but not with the overall survival rate (P>0.05); the disease-free survival rate of patients with high expression of CAP2 was poor, and the disease-free survival rate of patients with low expression of CAP2 was better. 【
Conclusion
】CAP2 is highly expressed in HCC tissues and cells, and is related to the prognosis of HCC. It provides biomarkers for the new diagnosis and prognosis of HCC.

WANG Zhi-qiang,SUN Peng, GAO Chun,et al
DOI: 10.3969/j.issn.1671-7171.2019.10.021
【
Objective
】 To investigate the relationship of chloride intracellularchannel 1expression (CLIC1) with invasion, metastasis and prognosis of gastric cancer (GC). 【
Methods
】 The GC tissues and adjacent tissues were obtained from 94 patients. The expression of CLIC1 mRNA and protein were detected. The correlation of CLIC1 protein expression with gastric cancer clinical characteristics was analyzed. Multivariate Cox proportional model was applied to reveal the influencing factors of poor prognosis of gastric cancer. 【
Results
】 The positive expression rate of CLIC1 protein in GC adjacent tissues was 25. 0% (23/94), which was significantly lower than that of the GC tissues 86% (81/94) (P<0.01) .The positive expression of CLIC1 was not correlated with age, gender, tumor metastasis, however, it was related with tumor differentiation, lymph node metastasis, depth of invasion and TNM stage (all P<0.05). The expression level of CLIC1 mRNA in gastric cancer tissues was significantly higher than that in adjacent normal tissues (P<0.05). The median survival time of patients with low expression of CLIC1 protein was over 29 months, which was significantly longer than that of patients with positive expression for 25 months ( P<0.05). The 5-year survival rate of patients with low expression of CLIC1 protein was 63.9%, which was significantly higher than 20.7% of patients with positive expression of CLIC1 protein (P<0.05). GC invasion depth, lymph node metastasis, TNM stage and CLIC1 protein expression were independent prognostic factors for gastric cancer survivals (P<0.01). 【
Conclusion
】 CLIC1 plays an important role in the genesis and development of gastric cancer. High expression of CLIC1 is closely correlated with the invasion and metastasis of GC.

Expression and Clinical Significance of Metastasis-associated in Colon Cancer-1 and KAI1/CD82 in Hepatocellular Carcinoma
WANG Chun-rong
DOI: 10.3969/j.issn.1671-7171.2019.10.023
【
Objective
】To explore the expression of MACC1 and KAI1/CD82 in hepatocellular carcinoma (HCC) patients , as well as the correlation with clinicopathological features. 【
Methods
】The expression levels of both MACC1 and KAI1/CD82 in 61 cases of HCC tissues and corresponding adjacent liver tissues were detected by immunohistochemistry, and the differences of MACC-1 and KAI1/CD82 protein expression between the two groups were compared. According to the clinical and pathological typing, the specimens of HCC radical resection group were grouped in order to reveal the difference of expression between the groups, and further explore the correlation between the expression of MACC-1 and KAI1/CD82 protein and clinical features. 【
Results
】Compared to adjacent tissues, MACC1 was significantly overexpressed in HCC tissues (P<0.05), while KAI1/CD82 was significantly down-expressed (P<0.05). The levels of MACC1 and KAI1/CD82 protein were correlated with tumor grade, lymph node metastasis, depth of invasion, and TNM stage (P<0.05). The overall survival time of patients with MACC1- or KAI1/CD82-positive HCC tumors was significantly longer than that of patients with MACC1 positive or KAI1 negative HCC (P<0.05). Importantly, multivariate analysis suggested that positive expression of either MACC1 or KAI1/CD82, as well as TNM stage, could be independent prognostic factors for overall survival in patients with HCC. 【
Conclusion
】 Abnormal expression of MACC1 and KAI1/CD82 plays an important role in the development of HCC. The combined detection of MACC1 and KAI1/CD82 is a biomarker for potential differential diagnosis of tumor metastasis and prognosis in HCC patients. MACC1 and KAI1/CD82 may represent promising metastatic and prognostic biomarkers, as well as potential therapeutic targets, for HCC.

】A total of 203 elderly HFPEF patients (observation group) admitted to our hospital from March 2016 to September 2017 were divided into two groups according to NYHA cardiac function grade Ⅱ (n=72), cardiac function grade Ⅲ (n=75) and cardiac function grade Ⅳ (n=56). Meanwhile, 50 healthy volunteers in the same period were selected as the control group. BUA, Hcy, NT-proBNP and LVEF were compared in each group. One year follow-up, according to the prognosis, patients were divided into poor prognosis group and good prognosis group. ROC curve was used to analyze the prognostic value of BUA and Hcy. 【
Results
】The levels of BUA, Hcy and NT-proBNP in the observation group were significantly higher than those in the control group, the difference was statistically significant (P<0.05); compared with levf, the difference was not statistically significant (P>0.05). The levels of BUA, Hcy and NT-proBNP were the highest in grade Ⅳ group, the second in grade Ⅲ group, and the lowest in grade Ⅱ Group (P<0.05). Pearson analysis showed that serum BUA and Hcy levels were positively correlated with NT-proBNP (r=0.784 and 0.732, respectively, P<0.05). 203 patients were hospitalized or died due to adverse mace events (poor prognosis group), including 27 cases of central colic, 19 cases of acute myocardial infarction, 13 cases of serious arrhythmia and 5 cases of death. The other 139 patients were included in the good prognosis group. The proportion of NYHA Ⅲ~Ⅳ, serum BUA, Hcy and NT-proBNP water in patients with good prognosis were lower than those in patients with poor prognosis (P<0.05).【
Conclusion
】The level of serum BUA and Hcy in HFPEF patients increases significantly, which is positively correlated with left ventricular function, and has a certain reference value for the diagnosis, severity and prognosis of HFPEF.
