Abstract:【objective】 To explore the effect of Jinhong Decoction on intestinal barrier function in rats infected with multidrug-resistant Pseudomonas aeruginosa (MDRPA). 【Method】 A total of 10 rats were randomly selected from 65 rats and given only 0.5 mL of physiological saline by gavage as the normal group; The remaining 55 rats were used to establish a model of intestinal MDRPA infection,and 50 rats were successfully modeled. They were randomly divided into model group,low dose group of Jinhong Decoction,medium dose group of Jinhong Decoction,high dose group of Jinhong Decoction,and piperacillin sodium tazobactam sodium (TZP) group,with 10 rats in each group. The low,medium,and high dose groups of Jinhong Decoction were gavaged with crude drug doses of 3.2 g/kg,6.3 g/kg,and 12.6 g/kg of Jinhong Decoction concentrated solution,while the TZP group was gavaged with TZP 70 mg/kg,with a dose volume of 10 mL/kg for each group,Continuously intervened for 2 weeks; The normal group and model group were given equal volume of physiological saline by gavage. The expression levels of intestinal barrier function,bacterial count of ileal contents,Toll like receptor 4 (TLR4) in colon tissue,myeloid differentiation factor 88 (MyD88),nuclear transcription factor κB p65(NF-κB p65)and p-NF-κB p65 protein of each group were compared. 【Results】 Compared with the normal group,the model group showed elevated levels of serum diamine oxidase (DAO),endotoxin (ET),and bacterial count in ileal contents (P<0.01); Compared with the model group,the levels of serum DAO and ET,as well as the bacterial count of ileal contents in the TZP group and each dose group of Jinhong Decoction decreased (P<0.05),and there was a dose-effect relationship between each dose group of Jinhong Decoction. Compared with the normal group,the expression levels of TLR4 and MyD88 proteins and p-NF-κB p65/NF-κB p65 in the model group increased (P<0.01); Compared with the model group,the expression levels of TLR4 and MyD88 proteins and p-NF-κB p65/NF-κB p65 in the TZP group and Jinhong Decoction at different doses decreased (P<0.05),and there was a dose-effect relationship between each dose group of Jinhong Decoction.【Conclusion】 Jinhong Decoction can improve the intestinal barrier function and reduce the bacterial count in the ileum of MDRPA infected rats,which may play a role by inhibiting the TLR4/MyD88 signaling pathway.
