丁苯酞对单侧输尿管梗阻大鼠肾间质纤维化的影响

摘要 目的 探讨丁苯酞对单侧输尿管梗阻(UUO)大鼠肾间质纤维化的影响及机制。方法 选取清洁级大鼠54只(雄性),建立UUO模型,随机分为3组,每组各18只:治疗组(A组) 和模型组(B组)于靠近肾盂段行左输尿管结扎,A组予丁苯酞(NBP)180 mg/kg/d灌胃;B组予等量生理盐水灌胃;假手术组(C组)仅分离输尿管,不行结扎,予等量生理盐水灌胃。各组在术后d2、d3、d7取大鼠肾脏和血液样品,测定肾脏脂质过氧化物反应标记物超氧化物歧化酶(T-SOD)和丙二醛(MDA)活性。结果 A组、B组、C组术后d3、d7与d2相比T-SOD水平降低,MDA水平升高(P<0.05)。三组术后d7分别与术后d3相比T-SOD水平降低,MDA水平升高(P<0.05)。B组术后d3、d7与A组相比T-SOD水平降低,MDA水平升高(P<0.05)。A组术后d3、d7与C组相比T-SOD水平降低,MDA水平升高(P<0.05)。A组、B组术后d3、d7分别与同组术后d2相比Ⅰ型胶原蛋白质水平均升高(P<0.05)。且B组术后d3、d7高于A组(P<0.05)。Ⅰ型胶原蛋白在C组有极少量表达,C组术后d2、d3、d7其水平无明显差异(P>0.05)。结论 NBP对UUO大鼠肾间质纤维化能够起到明显的调控作用,其原因可能与调节了MDA及T-SOD的活性具有一定的关系。

AbstractObjective To observe the effects of butylphthalide(NBP) on the renal interstitial fibrosis in rats with unilateral ureteral obstruction(UUO) and to explore the possible mechanism. Methods A total of 54 clean level male rats were selected to establish the UUO models. These rats were randomly divided into three groups with 18 in each group: group A and B with unilateral ureteral obstruction (left ureteral ligation in close proximity to renal pelvis) and group C without UUO (procedure mimic as control group). Rats in group A received NBP 180 mg/kg/d lavage, while rats in group B received equal dosage saline lavage. Rats in group C were only isolated the ureter with no ligation but give equal saline lavage as well. Rat kidney and blood samples were taken at postoperative d2, d3, and d7, and renal lipid peroxide reaction markers superoxide dismutase (T-SOD) and malondialdehyde (MDA) activities were measured. Results Compared to d2, the levels of T-SOD at postoperative d3 and d7 in group A, group B and group C decreased, while the level of MDA increased (P<0.05). T-SOD level was decreased and MDA level was increased in all three groups at d7 after surgery, if compared to d3 (P<0.05). Compared to group A, the level of T-SOD in group B was decreased, while the level of MDA was increased at postoperative d3 and d7 (P<0.05). Compared with group C, the level of T-SOD was decreased and the level of MDA was increased in group A at postoperative d3 and d7 (P<0.05). The levels of type I collagen protein in group A and group B at postoperative d3 and d7 were significantly higher than those in their matched group at postoperative d2 (P<0.05). Further, the levels of type I collagen protein in group B were greater than those in group A. The postoperative d3 and d7 of group B at postoperative d3 and d7 were higher than those of group A (P<0.05). Type I collagen was expressed in a small amount in group C, and there was no significant difference in the levels of d2, d3, and d7 in group C (P>0.05).Conclusion NBP plays a significant role in the regulation of renal interstitial fibrosis in rats in unilateral ureteral obstruction, which may be related to the regulation of the activity of MDA and T-SOD.
