Abstract:【Objective】 To investigate the effect of rubescene B combined with capecitabine on proliferation and invasion of breast cancer cells.【Method】 The breast cancer cell line 4T1was established and randomly divided into four groups: rubescene B group (group A), capecitabine group (group B), rubescene B + capecitabine group (group C) and blank control group (group D). MTT assay was used to detect the increment inhibition rate of breast cancer cell line 4T1 in each group. The effect of drugs on invasive ability of breast cancer cell line 4T1 was determined by Transwell chamber assay. RT-PCR method was used to detect the expression of beta-catenin, cyclin D1 and c-myc mRNA in breast cancer cell line 4T1. 【Results】 Treatment in group A, B and C all inhibited the proliferation of breast cancer 4T1 cells, and group C had the strongest inhibition, which was higher than the other groups (P<0.05). The invasiveness of breast cancer 4T1 cells was inhibited in group A, group B and group C. The invasiveness of breast cancer 4T1 cells in group C was the weakest, which was lower than that of the other three groups (P<0.05). The expression of beta-catenin, cyclin D1 and c-myc was significantly down-regulated by rubescene B combined with capecitabine (P<0.05). 【Conclusions】 Rubescene B combined with capecitabine can significantly inhibit the proliferation of breast cancer 4T1 cells, and inhibit cell invasion, which is related to the suppressing the Wnt/β-catenin signaling transduction pathway.
