Abstract:【Objective】To investigate the the cytotoxicity of oridonin on different malignant types of human bladder cancer cells. 【Methods】With mitomycin as a control, different concentrations of oridonin were treated to human bladder cancer T24 (invasive, G3) and 5637 (non-invasive, G2) cells. Cell proliferation inhibition was measured by MTT assay and IC50 values were calculated for comparison. Cell morphologic changes and apoptosis were observed by inverted microscope and fluorescence staining. 【Results】Oridonin and mitomycin inhibited cell proliferation and killed both T24 and 5637 cells, both showing time- and concentration-dependent effects. The proliferation inhibition and killing effects of oridonin at 64μM were significantly better than those of mitomycin at 150μM after treating T24 and 5637 cells for 12 h and 24 h. Compared with highly malignant invasive bladder cancer cells T24, low malignancy non-invasive bladder cancer cells 5637 were more sensitive to oridonin and mitomycin treatment. 【Conclusion】 Compared with mitomycin, oridonin induces apoptosis, inhibits proliferation, and kills bladder cancer T24 and 5637 cells to a greater extend. It is worthy of further research and development.
