Abstract:【Objective】 To investigate the expression of Periostin protein in esophageal squamous cell carcinoma and its correlation with the pathological features of patients. 【Methods】 A total of esophageal squamous cell carcinoma tissues (observation group) resected surgically in our hospital were collected, and the corresponding normal esophageal tissues (more than 5 cm from the cancer tissues) adjacent to the cancer were used as the control group. Immunohistochemistry, semi quantitative RT-PCR and Western blot were used to detect the expression of Periostin protein in esophageal squamous cell carcinoma tissues and normal esophageal tissues adjacent to the cancer. Kaplan-Meier survival curve Cox proportional hazard regression model was used to analyze the expression of Periostin protein and its correlation with the prognosis of patients with esophageal squamous cell carcinoma. 【Results】 The high expression rate of Periostin protein in the observation group was 63.6% (75/118), which was significantly higher than that in the control group [27.1% (32/118)]. The difference was statistically significant(χ2=31.614,P<0.001).The value of Periostin mRNA in esophageal squamous cell carcinoma tissues was higher than that in normal esophageal tissues (P<0.05). The value of Periostin/β-actin in normal tissues adjacent to esophageal carcinoma was lower than that in esophageal squamous cell carcinoma (P<0.05). The TNM stage, differentiation degree and lymph node metastasis of esophageal squamous cell carcinoma patients with different Periostin expression levels were statistically significant (P<0.05). The survival rate of patients with low expression of Periostin was significantly higher than that of patients with high expression of Periostin (P<0.05). Logistic multivariate regression analysis showed that the high expression of Priostin was a risk factor affecting the overall survival of patients with esophageal squamous cell carcinoma (P<0.05). 【Conclusion】 The expression of Periostin in esophageal squamous cell carcinoma is related to histological type, TNM stage and lymph node metastasis, which is of great significance in predicting the prognosis of esophageal squamous cell carcinoma.
