Abstract:【Objective】To investigate the effect and mechanism of zinc sulfate combined with aspirin in the treatment of acute myocardial infarction (AMI) rats. 【Methods】Healthy adult SD male rats were randomly divided into the sham operation group (normal saline perfusion in no AMI model rats), the model group (equal amount of normal saline gavage in AMI model rats), the zinc sulfate group [zinc sulfate 75 mg/(kg·d) gavage in AMI model rats], the aspirin group [aspirin75mg/(kg·d) gavage in AMI model rats], and the combined group [zinc sulfate and aspirin(2∶1) with total dose of 120mg/(kg·d) gavage in AMI model rats]. All rats kept continuous treatment for 7d. Rat lactate dehydrogenase (LDH), creatine kinase (CK), aspartate aminotransferase (AST), malondialdehyde (MDA), superoxide dismutase (SOD), gluten Glutathione peroxidase (GSH-Px), the maximum rate of increase of left ventricular pressure (+dp/dtmax), the maximum rate of increase of left ventricular pressure (-dp/dtmax), the interval time from the beginning of ventricle contraction to the occurrence of dp/dtmax (T-dp/dtmax), left ventricular end diastolic pressure (LVEDP), left ventricular systolic pressure (LVSP), and myocardial infarction area ratio (MIS%) were detected and compared among groups.【Results】The serum LDH, CK, and AST levels of the model group were significantly higher than those of the sham operation group (P<0.05); the serum LDH, CK, and AST levels of the combination group were significantly lower than those of the model group, zinc sulfate group, and aspirin group (P<0.05). The serum MDA level of the model group was significantly higher than that of the sham operation group (P<0.05), and the serum SOD and GSH-Px were significantly lower than that of the sham operation group (P<0.05). Serum MDA level in the combination group was lower than that in the model group, the zinc sulfate group and the aspirin group (P<0.05); while the serum levels of SOD and GSH-Px in the combination group were higher than those in the model group, the zinc sulfate group and the aspirin group (P<0.05). The measured values of +dp/dtmax, -dp/dtmax and LVSP of the model group were significantly lower than those of the sham operation group (P<0.05), and the measured values of +dp/dtmax, -dp/dtmax and LVSP of the combined group were significantly higher than those of the model group, the Zinc sulfate group and the aspirin group; the difference was statistically significant (P<0.05).The t-dp/dtmax, LVEDP and MIS% were significantly higher than the sham operation group. The values of t-dp/dtmax, LVEDP and MIS% in the combined group of rats were significantly lower than those in the model group, the zinc sulfate group, the aspirin group; and the difference was statistically significant (P<0.05). The myocardial tissues of rats in the sham operation group were arranged neatly, densely with clear structure. The cytoplasm was uniformly stained. The nucleus was located in the center of the cells. There was no inflammatory cell infiltration in the myocardium. The myocardial tissue of the AMI model group showed obvious structural damage, cell swelling, nuclear pyknosis and lysis, focal or flaky myocardial necrosis. The myocardial tissue of the zinc sulfate group and the aspirin group still showed obvious necrotic areas, but it was less than the model group. The myocardial tissue of the rats in the combination group showed only punctate necrosis or vacuole areas, and the myocardial pathology was significantly reduced. 【Conclusion】The results showed that zinc sulfate combined with aspirin treatment of AMI rats can significantly reduce myocardial infarct size, reduce myocardial enzymes and improve cardiac function in rats.
