Abstract:【Objective】To investigate the effects of PTX-3 gene expression on cell cycle regulation, proliferation, migration and invasion of endometrial cancer cells. 【Methods】By designing and synthesizing PTX3-specific siRNA, control plasmid and empty vector, HEC-1-A cells were transfected as PTX3-siRNA group, NC-siRNA group and Blank group, respectively. The expression of PTX3 mRNA in transfected cells was detected by RT-PCR. The cell cycle was analyzed by flow cytometry and the cell proliferation activity was measured by MTT assay. The scratch assay was used to reflect cell migration ability, and the Transwell chamber assay was applied to evaluate cell invasion ability. 【Results】The expression of PTX3 mRNA in PTX3-siRNA group was significantly lower than that in NC-siRNA group and Blank group (P<0.05). The proportion of cells in G1 phase in PTX3-siRNA group was significantly higher than that in NC-siRNA group and Blank group (P<0.05), while the proportion of S phase cells in PTX3-siRNA group was significantly lower than that of the NC-siRNA group and the Blank group (P<0.05). The cell proliferation ability, cell migration ability and the number of transmembrane cells in PTX3-siRNA group were significantly lower than those in the NC-siRNA group and the Blank group (P<0.05).【Conclusion】 Down-regulation of PTX3 gene expression may impede the progression of endometrial cancer cell cycle and inhibit cell proliferation, migration and invasion.
