Abstract：【Objective】To analyze the expression level and clinical value of CDKN2A gene in colon cancer based on TCGA database. 【Methods】In this study, TCGA RNA-seq data from the Tumor Immune Evaluation Database (TIMER) were collected to analyze the expression of CDKN2A in colon cancer and the impact of CDKN2A on the prognosis of colon cancer patients. The TIMER database was used to detect the infiltration of immune cells. The correlation between CDKN2A and clinical features was analyzed by R packet, and the protein interaction network information in CDKN2A gene was analyzed by STRING database. 【Results】CDKN2A gene was found to be overexpressed in 16 tumors, especially in colon cancer. The prognosis of colon cancer patients with higher CDKN2A expression level was worse, and the expression level of CDKN2A was significantly correlated with the T stage, N stage and pathological stage of the tumor (P<0.05). ROC curve analysis showed that CDKN2A was more effective in the diagnosis of colon cancer, and the area under the curve was 0.918. CDKN2A and immune checkpoint co-expression showed that CDKN2A was positively correlated with CD274, CTLA4, HAVCR2, LAG3, PDCD1, PDCD1LG2, SIGLEC15 and TIGIT (P<0.05). Spearman correlation analysis showed that CDKN2A expression was positively correlated with CCND1 expression (P<0.05). 【Conclusion】CDKN2A has high efficacy in the diagnosis and prognosis assessment of colon cancer patients, which is a potential target for the treatment of colon cancer, and has important significance in improving the diagnostic accuracy and prognosis assessment of colon cancer.
