Abstract:【Objective】To explore effects of ω-3 polyunsaturated fatty acids(ω-3PUFAs) on inflammatory bowel disease(IBD) and its mechanism. 【Methods】Peripheral blood samples were collected from 60 patients with active IBD, and peripheral mononuclear cells were isolated. They were randomly divided into three groups, 20 in each group.Respectively they were treated withω- 3PUFAs eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA) and saturated fatty acid stearic acid(SA) for 48 hours;In addition, 20 blood samples of healthy volunteers who underwent physical examination in our hospital in the same period were collected to obtain mononuclear cells. Cell culture was resuspended as the blank group. The morphological changes of cells in the four groups were observed after 48 hours; The number of CD4+CD25+FoxP3+ and CD4+CD25+CD127-T cells was detected by flow cytometry; The mRNA levels of CD80 and CD86 were detected by semi quantitative reverse transcription polymerase chain reaction(RT-PCR); Interleukin-12(IL-12), IL-10 and tumor necrosis factor-α(TNF-α)level in cell supernatant were measured by enzyme-linked immunosorbent assay(ELISA).【Results】The numbers of CD4+CD25+FoxP3+ and CD4+CD25+CD127-T cells in the blank group and the SA group were higher than those in the EPA group and the DHA group(P<0.05). The numbers of CD4+CD25+FoxP3+ and CD4+CD25+CD127-T cells in the SA group was lower than that in the blank group(P<0.05). The CD80 and CD86 mRNAs in supernatants in the blank group and the SA group were higher than those in the EPA group and the DHA group(P<0.05). The CD80 and CD86 mRNAs in supernatants in the SA group were lower than those in the blank group(P<0.05). The concentration of IL-12 and TNF-α in cell supernatant in the blank group and SA group was higher than that in the EPA group and the DHA group(P<0.05), while IL-10 concentration was lower than that in the EPA group and the DHA group(P<0.05). The concentration of IL-12 and TNF-α in cell supernatant in the SA group was lower than that in the blank group, while IL-10 concentration was higher than that in the blank group(P<0.05). There was no significant difference in the above indexes between the EPA group and the DHA group(P>0.05). 【Conclusion】 ω-3 PUFAs can regulate IBD immune disorder, maintain immune balance, reduce inflammatory response, promote the repair of damaged cell membrane, or may become a new direction of IBD treatment research.
