Relationship Study Between the Heat Shock Protein 70 (HSP70) and Activation of TLR4 Signaling Pathway in Peripheral Blood Mononuclear Cells In Unstable Angina Pectoris Patients

Abstract:【Objective】To evaluate relationship between the heat shock protein 70 (HSP70) and the activation of TLR4 signaling pathway in peripheral blood mononuclear cells in unstable angina pectoris (UAP) patients.【Methods】The clinical data of 30 patients with UAP and 30 cases with stable angina pectoris (SAP) who were admitted to our hospital from December 2014 to March 2016 were selected and divided into UAP group and SAP group . At the same time, 30 patients with suspected acute coronary syndrome (ACS) who were admitted to our hospital in the same period of time were selected as control group who showed a normal coronary artery after coronary angiography. The levels of HSP70 in peripheral blood serum and monocyte NF-κB p65 levels were detected by ELISA.Flow cytometry was used to detect the expression of TLR4 in peripheral blood mononuclear cells of each group, and the correlation of the parameters of each group was analyzed by order.【Results】Levels of HSP70, the positive rate of TLR4 expression in monocytes and the monocyte nuclear protein NF-κB p65 in the UAP group were higher than those in the SAP group and the control group, the difference was statistically significant (mean P<0.05); the positive expression rate of TLR4 in peripheral blood mononuclear cells of the SAP group was significantly higher than that of the control group, the difference was statistically significant (P<0.05). The HSP70 level in peripheral blood serum of the UAP group was significantly correlated with the positive expression rate of TLR4 and the nuclear protein NF-κB p65 (P<0.05) ; the positive expression rate of TLR4 was significantly correlated with the nuclear protein NF-κB p65 (P<0.05); There was no significant correlation on the above indexes between the SAP group and the control group (22) (P>0.05).【Conclusion】HSP70 may be involved in the activation of TLR4 signaling pathway in peripheral blood mononuclear cells of UAP patients, through the expression of monocyte TLR4 receptor.
