Abstract:【Objective】To investigate the expression of IgE high-affinity receptor γ subunit (FcεRIγ subunit) in the mononuclear cells of patients with atopic dermatitis (AD) and its regulation effect of acetylation level. 【Methods】 Twenty-three cases of AD patients (AD group) and 12 healthy volunteers (control group) were selected from the Dermatological Department of the Second Xiangya Hospital of Central South University, and the peripheral blood was collected from all subjects, and mononuclear cells were isolated, and the mononuclear cells from the control group was treated with the acetyltransferase inhibitor, trichostatin A (TSA), and the real-time fluorescence was used to detect the expression of FcεRIγ subunit in mononuclear cells, the expression of FcεRI on the surface of mononuclear cells was detected by flow cytometry; chromatin immunoprecipitation (CHIP) PCR was used to analyze the expression of histone H3 acetylation (ac-histone H3) in the promoter region of FcεRIγ subunit; Pearson correlation analysis was used to analyze the correlation between FcεRIγ subunit, FcεRI with FcεRI subunit ac-histone H3. 【Results】Compared with the control group, the expression of FcεRIγ subunit mRNA in the peripheral blood mononuclear cells of the control group +TSA treatment group and the AD group increased, the proportion of FcεRI cells and the average fluorescence intensity increased; and the FcεRIγ subunit ac-histone H3 mRNA expression increased (P<0.05). Pearson correlation analysis showed that FcεRIγ subunit and FcεRI were positively correlated with FcεRIγ subunit ac-histone H3. 【Conclusion】The FcεRIγ subunit gene of monocyte in AD patients can regulate the sequence acetylation, and promote the up-regulated expressions of FcεRIγ subunit and FcεRI on the surface of mononuclear cells, thus participating in the occurrence and development of AD.
