转染靶向UHRF1的siRNA对人宫颈癌HeLa细胞增殖和凋亡的影响

摘要 【目的】 探讨转染靶向泛素样含PHD和环指域1(UHRF1)的小干扰RNA(siRNA)对人宫颈癌HeLa细胞增殖和凋亡的影响。【方法】 将HeLa细胞随机分为对照组(未做处理)、空载组(转染阴性对照siRNA)和干扰组(转染靶向UHRF1的siRNA), RT-PCR和Western blot检测各组UHRF1 mRNA和蛋白的表达水平,并通过细胞增殖及毒性检测试剂盒(CCK-8)和Annexin V-FITC/PI染色流式细胞术检测干扰UHRF1基因表达后,对HeLa细胞增殖和凋亡的影响。【结果】 干扰组细胞中UHRF1 mRNA和蛋白表达水平显著低于对照组和空载组,其差异具有统计学意义(P<0.01);干扰组细胞的增殖受到抑制,细胞凋亡率升高,与对照组和空载组比较,其差异有统计学意义(P<0.01),但对照组和空载组比较差异均无统计学意义(P>0.05)。【结论】 通过转染siRNA靶向干扰HeLa细胞UHRF1表达,能够抑制HeLa细胞增殖,促进细胞凋亡。

Abstract【Objective】 To investigate the effects of ubiquitin-like containing PHD and ring finger 1 (UHRF1) gene silencing by specific small interfering RNA (siRNA) on the proliferation and apoptosis in human cervical cancer HeLa cells in vitro.【Methods】 HeLa cells were randomly divided into three groups: the untreated control group without transfection, the negative control group with scramble siRNA transfection and the silencing group which were transfected with the UHRF1 specific gene-targeted siRNA. The mRNA and protein levels of UHRF1 were detected by RT-PCR and western blot, respectively. The effects of UHRF1 gene silencing on the proliferation and apoptosis were analyzed by conducting CCK-8 assay and flow cytometry with Annexin V-FITC/PI staining. 【Results】 The RT-PCR and western blot results showed the mRNA and protein levels of UHRF1 were significantly decreased in HeLa cells after the transfection of UHRF1 gene-targeted siRNA (P<0.01). The proliferation was inhibited and apoptosis was triggered via silencing UHRF1 gene, and the difference between the silencing group and the untreated control group/ negative control group was statistically significant (P<0.01). However,There was no significant difference between the untreated control group and the negative control group (P>0.05). 【Conclusion】 The expression of UHRF1 gene was efficiently interfered by transfecting the UHRF1-targeted siRNA, which resulted in decreased proliferation and enhanced apoptosis in HeLa cells.
