Expression and Significance of miR-21,SP-A and SP-D in Serum and MRC-5 Cells of Patients with Pulmonary Fibrosis

Abstract  【Objective】To explore the significance and relationship of pulmonary fibrosis with miR-21 and SP-A,SP-D in serum and MRC-5 cells of pulmonary fibrosis patients.【Methods】From October 2020 to June 2021,30 patients with Pulmonary fibrosis (the pulmonary fibrosis group) and 30 healthy volunteers (the healthy group) were recruited in our hospital for the study. The expression levels of miR-21,SP-A,and SP-D in serum of the two groups were detected and compared. Meanwhile,MRC-5 cells were divided into an empty virus control group (transfected with meaningless miRNA lentivirus),a miR-21 overexpression group (transfected with miR-21 mimetic vector),and a miR-21 silencing group (transfected with inhibitor lentivirus vector). The expression levels of miR-21,SP-A,and SP-D mRNA in MRC-5 cells in each group were measured and compared. 【Results】 Serum levels of miR-21,SP-A and SP-D in the pulmonary fibrosis group were higher than those in the healthy group (P<0.05). In the cell experiment,levels of SP-A and SP-D in the miR-21 overexpression group were significantly higher than those in the empty virus control group (P<0.05). After silencing miR-21,the levels of SP-A and SP-D were significantly lower than those of the empty virus group (P<0.05). 【Conclusion】miR-21,SP-A and SP-D can be used as biomarkers of pulmonary fibrosis. And the expression of SP-A and SP-D decreases by inhibiting miR-21,which can inhibit the occurrence and development of pulmonary fibrosis.
