Abstract  【Objective】To investigate the effect of gabapentin combined with sodium hyaluronate on pain, β-endorphin and prostaglandin E2 levels in patients with knee osteoarthritis (KOA). 【Methods】A total of 86 patients with KOA treated in Yan'an people's Hospital from July 2019 to June 2021 were randomly divided into control group (given sodium hyaluronate) and observation group (given gabapentin + sodium hyaluronate), with 43 cases in each group. VAS score, knee function score and serumβ- Endorphin, prostaglandin E2 and their safety were compared between the two groups .【Results】Before treatment, there was no significant difference in VAS score, serum β-endorphin, prostaglandin E2 level and knee Lysholm score between the two groups (P>0.05). Compared with before treatment, the VAS scores of the two groups decreased after 1, 2 and 4 weeks of treatment, and the VAS scores of the observation group after 2 weeks of treatment and 4 weeks of treatment were lower than those of the control group (P<0.001). After treatment, the levels of the serum β- endorphins of the two groups of patients increased and the levels of prostaglandin E2 decreased in the observation group; The level of β- endorphin was higher than that in the control group (P<0.001), and the level of prostaglandin E2 was lower than that in the control group (P<0.001). After treatment, the knee Lysholm score of the two groups increased, and the knee Lysholm score of the observation group was higher than that of the control group (P<0.001). The total incidence of adverse reactions was similar between the two groups (χ2=0.453,P=0.501). 【Conclusion】 Gabapentin combined with sodium hyaluronate in the treatment of KOA patients can effectively relieve pain and regulate the levels ofβ- endorphin and prostaglandin E2, and improve knee functions, which is safe and reliable, and is worthy of clinical application.
