Effects of Leptin on Autophagy, VEGF Expression, Cell Proliferation, Migration and Lumen Formation of RF/6A Cells in Vitro

Abstract  【Objective】To investigate the effects of Leptin on autophagy, vascular endothelial growth factor (VEGF) expression, cell proliferation, migration and lumen formation of RF/6A cells in vitro. 【Methods】RF/6A cells were cultured with different concentrations of Leptin(50 ng/mL Leptin, 100 ng/mL Leptin, 200 ng/mL Leptin) for 24 hours. The expression levels of microtubule associated protein 1 light chain 3(LC3),Beclin-1, p62 and VEGF were detected by Western blotting. After the RF/6A cells were treated with Leptin combined with autophagy inhibitor 3-methyladenine (3-MA) and chloroquine (CQ), cell proliferation was detected by CCK8 method, cell migration was detected by Transwell method, and lumen formation was detected by Matrigel method. 【Results】Compared with the control group, different concentrations of Leptin could up regulate the expression of LC3, Beclin-1 and VEGF, and inhibit the expression of p62 (P<0.05); Compared with Leptin group, 3-MA and CQ could inhibit cell proliferation, reduce the number of migrating cells and the number of lumen formation (P<0.05). 【Conclusion】Leptin can activate autophagy in RF/6A cells, promote VEGF expression, promote cell proliferation, migration and lumen formation.
