| The Effect of MUC3A on the Proliferation of Extrahepatic Cholangiocarcinoma Cells |

Abstract 【Objective】To explore the effect of MUC3A on the proliferation of extrahepatic cholangiocarcinoma (EHCC) cells. 【Methods】QBC939 cells with high MUC3A expression were selected for cell transfection. Transient transfection was used, and shRNA interference sequences (#1 and #2) targeting the MUC3A gene were designed and synthesized. Recombinant lentiviral plasmids were packaged to produce viral solution, which was then used to infect EHCC QBC939 cells. The cells were divided into four groups for culture: untransfected cells (controll), empty virus-transfected cells (shRNA-Ctrl), and lentivirus-mediated shRNA-transfected cells (shRNA-MUC3A#1, shRNA-MUC3A#2). Western Blot (WB) was used to detect the silencing efficiency of shRNA on the MUC3A gene. Colony formation assay was used to detect the effect of MUC3A knockdown on the proliferation ability of QBC939 cells. 【Results】WB results showed that compared with the control group, the MUC3A expression in the shRNA-MUC3A#1 and shRNA-MUC3A#2 groups was decreased, and shRNA-MUC3A#2 had a better silencing effect, with statistically significant differences (all P<0.05). Colony formation assay results showed that 24 hours after transfection, silencing MUC3A significantly reduced the colony formation of QBC939 cells; both shRNA-MUC3A#1 and shRNA-MUC3A#2 could inhibit the proliferation of QBC939 cells, and shRNA-MUC3A#2 had a stronger inhibitory effect, with statistically significant differences (all P<0.05), indicating that MUC3A knockdown affects the proliferation ability of QBC939 cells. 【Conclusion】 MUC3A plays an important role in the proliferation of EHCC cells; knocking down the expression of MUC3A protein in EHCC cells can inhibit the proliferation of extrahepatic cholangiocarcinoma cells.
