Abstract:【Objective】Objective To investigate the expression and clinical significance of Na+/H+ exchanger regulatory factor 1 (NHERF1) and neurofibrin 2 (NF2) gene in colorectal cancer. 【Methods】 A total of 126 colorectal cancer patients diagnosed and treated in Ankang Central Hospital from March 2015 to June 2016 were selected. Samples of colorectal cancer tissues and normal tissues adjacent to cancer were collected. The positive expression of NHERF1 and NF2 in colorectal cancer tissues and normal tissues adjacent to cancer was detected by immunohistochemistry, and the relationship between the positive expression rate of NHERF1 and NF2 and the clinicopathological characteristics of colorectal cancer patients was analyzed; Cox proportional hazard regression model was used to analyze the prognostic risk factors of colorectal cancer patients. 【Results】The positive expression rate of NHERF1 in colorectal cancer was 74.6%, which was significantly higher than that in normal adjacent tissues (33.3%), and the positive expression rate of NF2 was 28.6%, which was significantly lower than that in normal adjacent tissues (77.8%, P<0.05). The positive rates of NHERF1 and NF2 were not correlated with age and sex (P>0.05); It was correlated with the maximum tumor diameter, TNM stage, depth of invasion, pathological grade, lymph node metastasis and recurrence (P<0.05). The 5-year survival rate of colorectal cancer patients with maximum tumor diameter ≥ 2 cm, TNM stage Ⅲ-Ⅳ, depth of invasion≥1 cm, lymphatic vascular invasion, high pathological grade, recurrence, NHERF1 positive, NF2 negative were significantly reduced (P<0.05). TNM stage Ⅲ-Ⅳ, high pathological grade, positive expression of NHERF1 and negative expression of NF2 were independent risk factors for the prognosis of colorectal cancer patients (P<0.05). 【Conclusion】The positive expression rate of NHERF1 protein in colorectal cancer is increased, while the positive expression of NF2 protein is decreased; NHERF1 positive expression and NF2 negative expression are independent risk factors for prognosis of colorectal cancer patients.
