Abstract:【Objective】 To investigate the expression and clinical significance of EB virus (EBV)-DNA, T-cadherin (T-cadherin) and proliferating nuclear antigen (PCNA) in the serum of patients with NPC. 【Methods】 From January 2013 to December 2015, 150 patients with advanced (Ⅲ-ⅣA) NPC were selected as NPC group, and 50 patients with chronic rhinitis (rhinitis group) and 50 healthy volunteers (control group) were selected as NPC group. The levels of EBV-DNA, T-cadherin mRNA and PCNA mRNA before and after treatment were detected by real-time fluorescent quantitative PCR, and the recurrence and metastasis were recorded after 2 years of follow-up, and the relationship between them and the above detection indexes was analyzed.【Results】 There were statistically significant differences in EBV-DNA positive rate, T-cadherin mRNA and PCNA mRNA between the three groups of the nasopharyngeal carcinoma group, the rhinitis group and the control group (P<0.05); compared with the treatment before treatment, the PCNA mRNA level of the nasopharyngeal carcinoma group was significantly decreased (P<0.05) and the T-cadherin mRNA was significantly increased (P<0.05). 124 cases of treatment sensitivity (TS) and 26 cases of TR were found in 150 patients with nasopharyngeal carcinoma; before treatment, EBV-DNA positive rate and PCNA mRNA in TR patients were higher than those in TS patients, while T-cadherin mRNA was lower than that in TS patients, the difference was statistically significant (P<0.05). Of 150 patients with nasopharyngeal carcinoma after radiotherapy , 35 of them had recurrence and metastasis, and the rate of recurrence and metastasis was 23.33% of which 3 had recurrence and 32 had distant metastasis.The positive rate of EBV-DNA in patients with metastasis was significantly higher than that in patients without recurrence and metastasis (P<0.05), T-cadherin mRNA in patients without recurrence and metastasis was higher than that in patients with recurrence and metastasis (P<0.05), PCNA mRNA in patients without recurrence and metastasis was lower than that in patients with recurrence and metastasis (P<0.05).【Conclusion】 The differential expression of EBV-DNA、T-cadherin and PCNA in the recurrence and metastasis of patients with advanced nasopharyngeal carcinoma before and after radiotherapy is expected to be an effective index for the evaluation of curative effect and prognosis.
