Expression of IL-22 of the from Peripheral Blood Mononuclear Cells in Patients with Rheumatic Heart Disease

Abstract:【Objective】To investigate the expression and significance of the interleukin-22 that secreted by the peripheral blood mononuclear cells from the patients with rheumatic heart disease (RHD) to provide theoretical basis for the clinical treatment. 【Methods】A total of 30 patients with RHD who underwent surgery from May 2016 to May 2017 were selected as the observation group and 30 healthy blood donors were selected as the control group. Peripheral blood mononuclear cells from patients with rheumatic heart disease and healthy donors were isolated by density gradient centrifugation. The content and gene expression of IL-22 in the culture medium were detected using ELISA and qRT-PCR, respectively. At the same time, the cellular morphology of peripheral blood mononuclear were observed by scanning electron microscope after co-cultured with the culture medium from patients with rheumatic heart disease . 【Results】The content and gene expression of IL-22 in the culture medium of patients with rheumatic heart disease were significantly higher than those in healthy donors. The differences were statistically significant(P<0.05). The morphology of peripheral blood mononuclear cells from healthy donors showed dramatically changed even collapsed and ruptured when they co-cultured with the secreted medium from RHD patients' mononuclear cells.. 【Conclusion】The over-expression of IL-22 of the peripheral blood mononuclear cells from patients with rheumatic heart disease plays a certain role in the occurrence and development of the disease RHD.
