Effect of 4-Methylumbelliferone on Dendritic Cells Derived from Peripheral Blood Mononuclear Cells

Abstract  【Objective】To investigate the effect of 4-methylumbelliferone (4MU) on monocyte derived dendritic cells (Mo-DCs) in human peripheral blood.【Methods】Human monocytes were obtained by density gradient centrifugation and cultured for 5 days by human granulocyte monocyte colony stimulating factor and interleukin-4, Mo-DCs were treated with different concentrations of 4mu, the activity of Mo-DCs in each group was detected by CCK-8 method, the immunophenotypic marker level of Mo-DCs in each group was detected by flow cytometry, and the IL-6 level of Mo-DCs in each group was detected by electrochemiluminescence method.【Results】 When the concentration of 4mu was 0.1~1 mmol/L, it has little effect on the activity of Mo-DCs. 4MU inhibited the expression of immunophenotypic markers CD40, CD83, CD80, HLA-DR and Mo-DCs secreting IL-6.【Conclusion】 4MU inhibits the maturation of Mo-DCs and reduces the secretion of IL-6, which provides a new idea for the prevention and treatment of infectious inflammation. 4MU may become a new drug for immune regulation.
