Effect of Parthenolide on Polarization of Macrophage Cell Line RAW264.7 |

Abstract 【Objective】 To investigate the effect of parthenolide (PAR) on the polarization typing of macrophage cell line RAW264.7. 【Methods】 The macrophage cell line RAW264.7 was divided into control group, model group and experimental group. The control group was given routine culture, the model group was added with 100μg/L lipopolysaccharide, the experimental group were added with low, medium and high concentrations of par (2.5μmol/L, 5μmol/L, 10μmol/L); after 7 days of continuous culture, the levels of TNF-α, MCP-1, TGF-β1 and IL-4 in RAW264.7 were detected by ELISA; flow cytometry was used to detect the expression of CD16/32 and CD206 on the surface of two kinds of cells; Western blot was used to detect the level of ApoE protein in three groups of par cells with different concentrations; real-time fluorescent PCR (RT-qPCR) was used to detect the levels of IL-10, Arg-1 and iNOS mRNA in three groups of cells.【Results】 The levels of TNF- α and MCP-1 in the experimental group were significantly lower than those in the control group and the model group (P<0.05), and the levels of TGF- β 1, IL-4 and apoE protein in the experimental group were significantly higher than those in the control group and the model group (P<0.05). The levels of TNF- α and MCP-1 in the experimental group decreased in turn, while the levels of TGF-β 1, IL-4 and apoE protein in the experimental group increased in turn, and the differences between the two groups were statistically significant (P<0.05). The M2/M1 of the model group was 0.47, significantly lower than 0.85 of the control group, while the M2/M1 of the experimental group was 0.61, 0.69 and 0.73, respectively, significantly higher than that of the model group (P<0.05); the M2/M1 of the low, medium and high concentrations increased in turn, with a statistically significant difference (P<0.05). Compared with the control group, the level of Arg-1 mRNA decreased significantly; the level of IL-10 and iNOS mRNA increased significantly (P<0.05); the level of IL-10 and iNOS mRNA in the high concentration of the experimental group was significantly lower than that in the low concentration and the middle concentration; the level of Arg-1 mRNA was significantly higher than that in the low concentration and the middle concentration, and the difference was statistically significant (P<0.05). 【Conclusion】 PAR may further induce RAW264.7 polarization from M1 pro-inflammatory phenotype to M2 anti-inflammatory phenotype.
