The Effect of Atorvastatin on Angiogenesis and VEGF Expression in Rats with Chronic Subdural Hematoma

Abstract  【Objective】To investigate the effect of atorvastatin on angiogenesis and the expression of vascular endothelial growth factor (VEGF) in chronic subdural hematoma (CSDH) rats. 【Methods】Thirty SPF grade SD rats were randomly divided into sham surgery group,model group,and atorvastatin group,with 10 rats in each group. After the successful establishment of the CSDH model,rats in the atorvastatin group were treated with atorvastatin by gavage for 7 consecutive days. The sham operated group and model group were gived with the same amount of physiological saline at the same time. Immunohistochemistry was used to detect microvascular density (MVD) in the outer membrane tissue of hematoma in rats of each group. Real time fluorescence quantitative PCR was used to detect the expression level of VEGF mRNA in the brain tissue of rats of each group. Immunoblotting was used to detect the changes in VEGF protein levels in the brain tissue of rats of each group. 【Results】As time passed,the volume of intracranial hematoma in the model group and atorvastatin group of rats was lower than that on day 3,and the atorvastatin group was lower than the model group,with a statistically significant difference (P<0.05). Compared with the sham surgery group,the levels of MVD-CD34 in the brain tissue of the model group and atorvastatin group were both increased (P<0.05),while the atorvastatin group was lower than the model group (P<0.05). Compared with the sham surgery group,the levels of VEGF mRNA in the brain tissue of the model group and atorvastatin group were both increased,while the atorvastatin group was lower than the model group,with a statistically significant difference (P<0.05). Compared with the sham surgery group,the levels of VEGF protein in the brain tissue of the model group and atorvastatin group were both increased,while the atorvastatin group was lower than the model group (P<0.05). 【Conclusion】Atorvastatin may inhibit neovascularization in brain tissue by down regulating VEGF expression,thereby inhibiting the occurrence and development of CSDH.
