Correlation and Clinical Significance of miR-130a and RUNX3 Expression in Esophageal Squamous Cell Carcinoma

Abstract  【Objective】To investigate the correlation and clinical significance of the expression of miR-130a and recombinant human Runt-related transcription factor 3 (RUNX3) in esophageal squamous cell carcinoma. 【Methods】A total of 84 patients with esophageal squamous cell carcinoma who were treated in our hospital were collected. The cancer tissues and the corresponding adjacent tissues of patients at the time of surgery were taken for exam. The qRT-PCR was performed to detect the expression level of miR-130a in tissues; Western blot and immunohistochemical methods were performed to detect the expression of RUNX3 protein in tissues. All postoperative patients were followed up to investigate the effects of miR-130a and RUNX3 expression on the prognosis of patients with esophageal squamous cell carcinoma. 【Results】Compared with adjacent tissues,the expression level of miR-130a in esophageal squamous cell carcinoma tissues was higher (P<0.05),while the expression level of RUNX3 protein was lower (P<0.05). The expression levels of miR-130a and RUNX3 protein in esophageal squamous cell carcinoma were negatively correlated (P<0.05). The protein expression levels of miR-130a and RUNX3 were obviously correlated with T stage,cancer differentiation and lymph node metastasis (P<0.05). Kaplan-Meier survival curve analysis showed that the median survival time of patients with esophageal squamous cell carcinoma of high miR-130a expression was obviously shorter than that of patients with low miR-130a expression (P<0.05).Also the result showed that the median survival time of patients with esophageal squamous cell carcinoma with low RUNX3 expression was obviously shorter than that of patients with high RUNX3 expression (P<0.05). Multivariate Cox analysis showed that T staging (T3-T4),lymph node metastasis,high expression of miR-130a,and low expression of RUNX3 were independent risk factors for poor prognosis in patients with esophageal squamous cell carcinoma (P<0.05). 【Conclusion】The expression level of miR-130a is up-regulated and the expression level of RUNX3 protein is down-regulated in esophageal squamous cell carcinoma. The two are closely related to the poor prognosis of patients with esophageal squamous cell carcinoma. Patients with esophageal squamous cell carcinoma with high expression of miR-130a and low expression of RUNX3 may have a higher risk of poor prognosis.
