Expression of miR-155 HG in Peripheral Blood Mononuclear Cells of Patients with Psoriasis Vulgaris and its Relationship with Th17/treg Cell Balance

Abstract  【Objective】 To investigate the expression level of microRNA-155 host gene (miR-155 HG) in peripheral blood mononuclear cells (PBMC) of patients with psoriasis vulgaris and its relationship with the balance of helper T cell 17/regulatory T lymphocyte (Th17/treg) cells.【Methods】 A total of 92 blood samples from patients with psoriasis vulgaris treated in the Yulin Second Hospital from October 2020 to October 2021 was selected as disease group, and 92 blood samples from healthy people in the same period were selected as control group. The expression levels of miR-155, Hg and miR-155 in PBMC were detected by real-time quantitative PCR (RT-PCR), and the proportion of Th17 and treg cells was detected by flow cytometry, Th17/treg valu was calculated; Serum interleukin-6 (IL-6) and transforming growth factorβ1(TGF-1) were detected by enzyme-linked immunosorbent assay (ELISA) ; The levels of miR-155 and miR-155 HG in PBMC and their relationship with Th17/treg value, IL-6 and TGF-β1 were analyzed by Pearson method.【Results】Compared with the control group, the levels of miR-155, miR-155 HG, IL-6 and Th17/treg in PMBC in the disease group were significantly increased (P<0.05), and serum TGF-β1 decreased (P<0.05). Of patients in the disease group, the level of miR-155 HG in PBMC was positively correlated with that of miR-155 (r=0.573,P=0.000); The level of miR-155 HG was positively correlated with Th17/treg value and IL-6 level (r=0.482, 0.512,P=0.000), and was negatively correlated TGF-β1 (r=-0.596,P=0.000); The level of miR-155 was positively correlated with Th17/treg value and IL-6 level (r=0.694, 0.488,P=0.000), and was negatively correlated with TGF-β1 level (r=-0.509, P=0.000). 【Conclusion】The level of miR-155 HG in PBMC of patients with psoriasis vulgaris is up-regulated, which may be related to the imbalance of secreting miR-155 and Th17/treg cells.
