													     		                            						                            																	    Abstract  【Objective】  To investigate the effects of ubiquitin-like containing PHD and ring finger 1 (UHRF1) gene silencing by specific small interfering RNA (siRNA) on the proliferation and apoptosis in human cervical cancer HeLa cells in vitro.【Methods】  HeLa cells were randomly divided into three groups: the untreated control group without transfection, the negative control group with scramble siRNA transfection and the silencing group which were transfected with the UHRF1 specific gene-targeted siRNA. The mRNA and protein levels of UHRF1 were detected by RT-PCR and western blot, respectively. The effects of UHRF1 gene silencing on the proliferation and apoptosis were analyzed by conducting CCK-8 assay and flow cytometry with Annexin V-FITC/PI staining. 【Results】  The RT-PCR and western blot results showed the mRNA and protein levels of UHRF1 were significantly decreased in HeLa cells after the transfection of UHRF1 gene-targeted siRNA (P<0.01). The proliferation was inhibited and apoptosis was triggered via silencing UHRF1 gene, and the difference between the silencing group and the untreated control group/ negative control group was statistically significant (P<0.01). However,There was no significant difference between the untreated control group and the negative control group (P>0.05). 【Conclusion】  The expression of UHRF1 gene was efficiently interfered by transfecting the UHRF1-targeted siRNA, which resulted in decreased proliferation and enhanced apoptosis in HeLa cells.
