Expression and Significance of Integrin α 5 and caspase 3 in Hepatocellular Carcinoma

Abstract  【Objective】 To observe and analyze the expression and clinical significance of integrin α 5 and aspartate protease 3 containing cysteine (caspase 3) in patients with hepatocellular carcinoma(HCC). 【Methods】 Paraffin-embedded specimens of 40 HCC cancer and paracancerous tissues from January to December 2015 underwent surgical resection in our hospital HCC were collected. Immunohistochemical examination of integrin α 5 and caspase 3 expression was performed. Also, fresh HCC tissues and corresponding paracancerous tissues were collected from 30 HCC patients who underwent surgical resection between January to December, 2016. Fluorescence quantitative RT-PCR and Western blotting were used to detect integrin α 5 and caspase 3 expression.【Results】The expression rate of integrin α 5 in HCC was significantly higher than that in adjacent tissues, while the expression of caspase 3 in HCC was lower than that in paracancerous tissue (P<0.05). The expression of integrin α 5 protein in 40 patients with HCC was closely related to TNM stage and tumor metastasis (P<0.05), while caspase 3 protein expression was related to HBsAg,TNM stage and tumor metastasis (P<0.05). There was a negative correlation between integrin α 5 and caspase 3 expression in HCC (P<0.05). The relative expression of integrin α 5 mRNA in HCC was significantly higher than that in adjacent tissues.The caspase 3 mRNA in the liver cancerous tissue was significantly lower than that in the paracancerous tissue (P<0.05). 【Conclusion】The negative correlation between integrin α 5 and caspase 3 may be involved in the apoptosis of hepatoma cells. However, the mechanism of integrin α 5 on apoptosis of hepatoma cells requires further study.
