miR-202在膀胱癌细胞中的表达水平及其对膀胱癌细胞增殖、凋亡和侵袭的影响

Abstract 【Objective】To investigate the expression of miR-202 in bladder cancer cells and its effect on proliferation, apoptosis and invasion of bladder cancer cells. 【Methods】The relative expression of miR-202 in bladder cells was detected by real-time fluorescence quantitative polymerase chain reaction (qRT-PCR); The miR-202 and miR-NC plasmids were transfected into UM-UC-3 cells by transfection technology and set as miR-202 group and NC group. Cell proliferation test, cell invasion test and apoptosis test were used to detect the proliferation, migration, invasion and apoptosis of cells in the two groups respectively. The volume and weight of transplanted tumors in the two groups were compared. Western blot test was used to detect the expression of PIK3CA protein in the cells of the two groups. 【Results】The relative expression level of miR-202 in bladder cancer cells was lower than that in normal bladder epithelial cells (P<0.05). Compared with NC group, the OD value, migration and number of invasive cells in the miR-202 group decreased, and the apoptosis rate increased, with significant difference (all P<0.05). The volume and weight of transplanted tumor in the miR-202 group were lower than those in the NC group, and the difference was statistically significant. The expression of PIK3CA protein in the miR-202 group was lower than that in the NC group, and the difference was statistically significant. 【Conclusion】miR-202 may inhibit the proliferation, migration, invasion and induce apoptosis of bladder cancer cells by inhibiting the expression of PIK3CA protein.
