Abstract:【Objective】To detect the levels of interleukin-35 (IL-35) and interleukin-17 (IL-17) in serum and aqueous humor of patients with diabetic retinopathy (DR), and to explore their clinical significance in the pathogenesis of DR. 【Methods】A total of 80 DR patients (80 eyes) who underwent cataract phacoemulsification in our hospital from March 2019 to March 2021 were selected as the study objects, while 65 elderly cataract patients (65 eyes) received cataract phacoemulsification in the same period were selected as the control group. The general clinical data of patients were collected and compared between the two groups. Enzyme-linked immunosorbent assay (ELISA) was used to determine the levels of IL-35 and IL-17 in serum and aqueous humor of subjects. Pearson correlation was used to analyze the correlation of IL-35 and IL-17 in serum and aqueous humor. The diagnostic values of IL-35 and IL-17 in serum and aqueous humor alone and their combination in DR were evaluated by receiver operating characteristic curve (ROC). 【Results】Compared with the control group, the levels of IL-35 in serum and aqueous humor of DR group were significantly lower (P<0.05), while the levels of IL-17 were significantly higher (P<0.05). Furthermore, the levels of IL-35 and IL-17 in serum of DR patients were lower than those in aqueous humor (P<0.05). Results showed that the levels of IL-35 and IL-17 in serum and aqueous humor of DR patients were negatively correlated (P<0.05). The area under curve (AUC) of IL-35, IL-17 and their combination in serum in the diagnosis of DR was 0.843, 0.863 and 0.957 respectively. The AUC of the combined detection of IL-35 and IL-17 in the diagnosis of DR was significantly higher than that of the single diagnosis of IL-35 and IL-17 in the serum (P<0.05). The AUC of IL-35 and IL-17 levels in aqueous humor and their combined detection for the diagnosis of DR was 0.830, 0.882 and 0.963 respectively. The AUC of the combined detection of both IL-35 and IL-17 in the diagnosis of DR was significantly higher than that of the independent diagnosis of IL-35 and IL-17 levels in aqueous humor (P<0.05). 【Conclusion】The expression levels of IL-35 in serum and aqueous humor of DR patients are low, and the expression levels of IL-17 are high in serum and aqueous humor of DR patients, which have certain diagnostic effects on DR. However, the method of serum sample collection is simple and more practical.
