Abstract:【Objective】To study the effect of hyaluronic acid (HA) mediated nano silver targeted liposomes on burn wound healing, and to explore the ratio of HA to Ag sulfadiazine.【Methods】The formulation includes 100% sulfadiazine silver (control formulation), 95% sulfadiazine silver + 5% hyaluronic acid (formulation a), 90% sulfadiazine silver + 10% hyaluronic acid (formulation b). 54 SD rats with deep second degree burn were randomly divided into control group, experimental group A and experimental group B, with 18 rats in each group. Six rats in each group were killed on d7 and d14 after the application of control formula, formula A and formula B respectively. The changes of hydroxyproline, collagen ratio and DNA proliferation cycle of dermal cells were measured.The time of wound healing and the area of residual wound were observed.【Results】The content of hydroxyproline and the proportion of type Ⅲ/Ⅰ collagen in the wound tissue of group d7 were higher than that of group d3. The content of hydroxyproline and the proportion of type Ⅲ/Ⅰ collagen in the wound tissue of group A were higher than that of group A. the content of hydroxyproline and the proportion of type Ⅲ/Ⅰ collagen in the wound tissue of group B were higher than that of group A (P<0.05). The DNA in G0/G1 phase of dermal cells in d5, d7, d9 and d11 group was significantly lower than that in d3 group, and that in S phase was significantly higher than that in d3 group (P<0.05). The proportion of DNA in G0/G1 phase of d5, d7 and d9 dermal cells in the group A was lower than that in the control group, and the proportion of DNA in G0/G1 phase of d11 dermal cells in the group B was higher than that in the group A; the proportion of DNA in G0/G1 phase of d5 and d7 dermal cells in the group B was lower than that in the group A, and the proportion of DNA in G0/G1 phase of d9 and d11 dermal cells in the group B was higher than that in the group A (P<0.05). The area of residual wound in the group A was lower than that in the control group. The area of residual wound in the group B was lower than that in the group A (P<0.05). The average healing time of the control group was (14.46±1.17) days, higher than that of the experimental group A (12.87±0.75) days, and the average healing time of the experimental group A was higher than that of the experimental group B (11.87± 0.55 days), the difference was statistically significant (P<0.05). 【Conclusion】Hyaluronic acid mediated nano silver targeting liposomes can effectively promote wound healing,and the combination of 90% sulfadiazine and 10% hyaluronic acid is ideal for promoting wound healing.
