Effect of TUSC3 on Proliferation, Migration and Invasion of Human Hepatoma Cells

Abstract  【Objective】To investigate the effect of tumor suppressor candidate 3 (TUSC3) on proliferation, migration and invasion of hepatoma cells(SMMC-7721). 【Methods】TUSC3 overexpression vector was transfected into SMMC-7721 cells, and TUSC3 mRNA and protein expression were detected by Real-time PCR and Western blot to verify the transfection efficiency of plasmid. MTT was used to detect cell proliferation at 24 h, 48 h, 72 h and 96 h after transfection. The cell migration ability was detected by wound healing and the invasion ability was detected by Transwell chamber. 【Results】TUSC3 was overexpressed in SMMC-7721 successfully. The results of MTT showed that the cell proliferation in TUSC3-overexpression group was significantly lower than that in empty vector group (P<0.01). The results of the wound healing showed that the cell migration in the TUSC3 overexpression transfection group was also significantly decreased (P<0.05). Similar to wound healing, the results of Transwell also showed that the invasive of hepatoma cells decreased after TUSC3 overexpression (P<0.01). 【Conclusion】 TUSC3 could inhibit proliferation, migration and invasion of hepatoma cells cells, and may play a tumor suppressing role in the development of hepatic carcinoma.
