													     		                            						                            																	    Abstract  【Objective】  To evaluate the expression of miR-423-5p in ovarian cancer tissues and investigate the effect of miR-423-5p on the proliferation of ovarian cancer cells. 【Methods】  The expression of miR-423-5p in 25 pairs of ovarian cancer and adjacent tissue was determined by real-time PCR. The expression of miR-423-5p in four ovarian cancer cell lines (SK-OV-3, A2780, COC1 and OVCAR3) was measured as well. The miR-423-5p-low-expressed cells were transfected with miR-423-5p mimic or NC mimic and the miR-423-5p-high-expressed cells were transfected with miR-423-5p inhibitor or NC inhibitor. Cell proliferation activities were examined by MTT assay. The expression level of ING-4 (growth inhibitory gene 4) was measured by real-time PCR. Dual-Luciferase reporter assay was also used to examine whether or not ING-4 was a direct target of miR-423-5p. 【Results】  The level of miR-423-5p was significantly increased in ovarian cancer tissues compared to cancer-adjacent tissue (P<0.05). A2780 cell showed the lowest level and OVCAR3 cell showed the highest level of miR-423-5p among the four ovarian cancer cell lines.  A2780 cells transfected with miR-423-5p mimic significantly increased cell proliferation activity while also remarkably inhibited the expression of ING-4 compared to NC mimic transfected A2780 cells. In contrast,  OVCAR3 cells after transfection with miR-423-5p inhibitor significantly inhibited cell proliferation and increased ING-4 level when compared to NC inhibitor transfected cells. Dual-Luciferase reporter assay showed that when compared to wt-ING-4 3'UTR+NC mimic group, the relative luciferase activity was significantly increased in wt-ING-4 3'UTR+miR-423-5p mimic group (P<0.05). However, there were unchanged in mut-ING-4 3'UTR+NC mimic group and mut-ING-4 3'UTR+miR-423-5p mimic group. 【Conclusion】  miR-423-5p is closely related to the occurrence and development of ovarian cancer. It may may be a novel target in the prevention and treatment of ovarian cancer.
