| Effect of Cofilin-1 Vverexpression on the Invasion, Proliferation and Apoptosis of Human Glioma Cells |

Abstract 【Objective】 To analyze the expression level of Cofilin-1 in glioma cell lines and explore its effects on proliferation, apoptosis and invasion of glioma cells. 【Methods】 Cofilin-1 overexpression plasmid was constructed and transfected into human glioma cell line U-87MG cells. The transfection efficiency of Cofilin-1 was detected by Real-time PCR and Western blot. The proliferation of U-87MG was assessed by MTT assay; Transwell assay was used to detect cell invasion; Contents of VEGF and MMP-9 in cell culture supernatant were measured by ELISA; Western blot was used to detect the protein levels of bax, Bcl-2, AKT, and p-AKT. 【Results】 Cofilin-1 overexpression plasmids were constructed by gene cloning technique and transfected into human glioma cell line U-87MG cell line. Real-time PCR and Western blot were used to test the effect of Cofilin-1 transfection. The levels of vascular endothelial growth factor (VEGFA) and matrix metalloproteinase-9 (MMP-9) in the supernatant of cultured cells were determined by ELISA assay. The expressions of bax, Bcl-2, AKT and p-AKT proteins were detected by Western blot. MTT assay was used to detect cell proliferation and transwell chamber assay was used to detect cell invasion ability.ELISA assay was used to detect vascular endothelial growth factor (VEGFA) and matrix metalloproteinase-9 (MMP-9) in supernatant of cell culture. The expression of apoptosis-related proteins (bax and Bcl-2, AKT and p-AKT) was detected by Western blot.【Conclusion】 Overexpression of Cofilin-1 can promote the proliferation and invasion of glioma cells. The mechanism may be that the activation of AKT pathway may play a role in Cofilin-1, which may become a potential therapeutic target for glioma.
