Expression and Significance of IL-17 mRNA and IFN-γ mRNA in Rat Models of Endotoxin Induced Uveitis |

Abstract 【Objective】To analyze the expression of interleukin-17 (IL-17) and interferon gamma (IFN- γ) messenger RNA (mRNA) in eyes of rats with endotoxin induced uveitis.【Methods】Forty SD rats were randomly divided into the blank control group (n=10) and the model group (n=30). Rats in the model group were treated with endotoxin to induce uveitis, and 5 rats were sacrificed at 6 h, 12 h, 18 h, 24 h, 48 h, 60 h, respectively;the control group was injected with phosphate buffered saline (PBS), and 10 rats were sacrificed after 24h. Uveal retina of all of them were taken from the both eyes. Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the levels of IL-17 mRNA and IFN- γ mRNA of ocular tissues in the two groups. The inflammation of the eye tissue was observed under microscope and scored according to relevant criteria.【Results】① The ocular inflammation score in the model group at different time was higher than that in blank control group (P<0.05);in the model group, the inflammatory scores were increased successively after 6 h, 12 h and 18 h (P<0.05), and reached the peak at 18 h, then decreased successively at 24 h, 48 h, 60 h(P<0.05). ② The IL-17 mRNA and IFN-γ mRNA levels in the model group at different times were higher than those in the blank control group (P<0.05); levels of IL-17mRNA and IFN-γ mRNA in the model group at 6 h, 12 h, 18 h increased successively (P<0.05), and reached the peak at 18 h , then successively decreased at 24 h, 48 h, 60 h (P<0.05).【Conclusion】The expression levels of IL-17 mRNA and IFN-γ mRNA in the ocular tissues of uveitis rats are up regulated, which are consistent with the extent of lesions; IL-17, IFN-γ may be involved in the pathogenesis of uveitis.
