Effects of Honokiol on Inhibition of Airway Inflammation During Acute Asthma and its Possible Mechanism |

Abstract [Objective]To explore the effects of honokiol on inhibition of airway inflammation during acute asth‐ma and analyze its possible mechanism .[Methods]A murine model of acute asthma was established by stimulation and induction with VOA .After a treatment of honokiol ,total cells ,neutrophils ,lymphocytes ,macrophages and eosino‐phils cells were counted .And the levels of tumor necrosis factor‐alpha (TNF‐α) ,interleukin‐4 (IL‐4) ,immunoglob‐ulin E (IgE) and Eotaxin were detected by enzyme‐linked immunosorbent assay (ELISA) in bronchoalveolar lavage fluid .Meanwhile ,lung tissue was sectioned for hematoxylin & eosin staining to analyze the histopathological chan‐ges .Moreover ,the expressionx of NF‐κBp65 and I‐κb were detected by Western blot .[Results]Honokiol effectively reduced the total cell number in bronchoalveolar lavage fluid (30976 .00 ± 3865 .54 vs 64861 .67 ± 7613 .98) while de‐creased the counts of neutrophils ,lymphocytes ,macrophages and eosinophils count and the differences were statisti‐cally significant ( P < 0 .05) .The results of ELISA showed that honokiol effectively inhibited the expressions of TNF‐α,IL‐4 ,IgE and Eotaxin .And the differences were statistically significant ( P<0 .01) .The results of pathol‐ogy showed that the area of tissue damage and inflammatory cells were effectively alleviated after a treatment of hono‐kiol as compared with model group .As compared with model group ,the expressions of NF‐κBp65 and I‐κb decreased after a treatment of honokiol and the difference was statistically significant ( P <0 .05) .[Conclusion]Honokiol can significantly inhibit the inflammation and lung tissue damage induced by acute asthma .And its possible mechanism is through an inhibition of NF‐κB activation .
