Abstract [Obj ective]To explore the effect of the up-regulation of transcription factor SOX2 on cell prolif-eration and invasion of cervical cancer cells.[Methods]The specific short hairpin(shRNA)SOX2 interference vector was transfected into cervical squamous cell SiHa in order to obtain stable transfection cell line(SOX2 in-terference group).MTT assay was used to detect the change of cell proliferation.The difference of cell inva-sion was detected by using Transwell invasion cabin.Western Blot was used to detect the expressions of SOX2,MMP2,MMP9 and epithelial-mesenchymal transition associated factors,which were compared with those in negative transfection control group and blank control group.[Results]MTT result showed that cell proliferation in SOX2 interference group was significantly reduced in comparison with negative transfection control group and blank control group.Transwell result showed that cell-penetrating number was significantly reduced and cell invasion was decreased(P<0.05).Western Blot result showed that SOX2 was up regulated markedly,and the expressions of MMP2,MMP9 were decreased,but the protein level of E-cadherin was in-creased and the expression of Vimentin was decreased(P<0.05).There was no significant difference in all indicators between negative transfection control and blank control group.[Conclusion]Suppression of SOX2 expression can effectively inhibit proliferation and invasion of cervical cancer cell in vitro,which may be related to the inhibition of epithelial-mesenchymal transition processes.
