Study on the Correlation Between-857C/T and -1031C/T Singe Nucleotide Polymorphism of Tumor Necrosis Factor-α Gene and Gestational Diabetes Mellitus |

Abstract [Objective]To explore the association between singe nucleotide polymorphism(SNP)of two lo-ci -857C/T and -1031C/T of tumor necrosis factor-α(TNF-α)gene and gestational diabetes mellitus (GDM).[Methods]Totally 50 normal pregnant women(control group)and 50 pregnant women with GDM (group GDM)in Beijing aerospace general hospital from March 2012 to Feb 2013 were selected.Peripheral ve-nous blood was collected to extract DNA.Polymerase chain reaction(PCR)restriction enzyme fragment length was used to analyze -857C/T and -1031C/T.The sequencing was performed for the confirmation.Chi-square test was applied to confirm the Hardy-Weinberg equilibrium in genotype distribution.Statistical meth-ods involving t test and chi-square test were conducted to evaluate the statistical significance in all data.[Re-sults]There was significant difference in genotype distribution and allele frequency of-857C/T of TNF-αgene between GDM group and control group(P<0.05).The proportion of CT genotype and the frequency of allele genotype T in GDM group was significant higher than that in normal pregnant group(P<0.05).However, there was no significant difference in genotype distribution and allele frequency of -1031 C/T locus between GDM group and control group(P >0.05).[Conclusion]The -857C/T allele gene polymorphism of TNF-αgene is associated with the pathogenesis of GDM.Pregnant women with T locus are susceptible to have GDM. But -1031C/T polymorphism is not related with GDM.
