Abstract [Objective] To explore the expressions of tumor suppressor gene PTEN and DNA mismatch repair gene hMSH2 in young women with endometrial carcinoma and their correlation .[Methods]The endometrial specimens of 66 women(≤45 years old) undergoing total hysterectomy for uterine diseases were collected .Among them ,there were 42 cases of endometrial carcinoma ,6 cases of atypical endometrial hyperplasia and 18 cases of normal endometri-um .The expressions of PTEN and hMSH2 in the tissues of endometrium were detected by immunohistochemical SP method .[Results] The expression rates of PTEN in normal endometrium ,atypical endometrial hyperplasia and endo-metrial carcinoma were decreased gradually ,and the absence rates were 16 .67% ,66 .67% and 85 .71% respectively , and there was significant difference ( P < 0 .01) .The expression rates of hMSH2 were 44 .44% ,33 .33% and 73 .81% ,respectively ,and there was significance difference among groups ( P <0 .05) .The expressions of PTEN and hMSH2 had no correlation with clinical stage ,histological grade ,myometrial invasion depth and lymphatic metas-tasis whether or not(rs=0 .271 ,P>0 .05) .There was no significant correlation between hMSH2 and PTEN in en-dometrial carcinoma( P < 0 .05) .The expression of hMSH2 was discordant with the expression of PTEN (χ2 =23 .04 ,P=0 .000) .[Conclusion] The combined detection of PTEN and hMSH2 is helpful for the early diagnosis and can infer the prognosis .
